miR-222在大鼠骨骼肌損傷再修復(fù)中的表達(dá)

【摘要】:為分析miR-222在骨骼肌損傷修復(fù)中的表達(dá)水平,本研究選取跑臺(tái)誘導(dǎo)骨骼肌損傷模型,參照Armstrong的實(shí)驗(yàn)動(dòng)物模型,選取2月齡健康雄性SD大鼠,建立股直肌損傷模型,在下坡跑運(yùn)動(dòng)結(jié)束后的不同時(shí)刻取材,對(duì)血清CK水平進(jìn)行檢測(cè),對(duì)股直肌冰凍切片(HE染色;橫切)進(jìn)行組織形態(tài)學(xué)分析,采用RT-PCR試驗(yàn)對(duì)miR-222水平進(jìn)行檢測(cè)。血清CK在股直肌運(yùn)動(dòng)后損傷過程中的表達(dá)水平持續(xù)增高;HE染色發(fā)現(xiàn):安靜對(duì)照組顯示肌纖維排列緊密規(guī)律,大小均勻,呈多邊形;與對(duì)照組相比,運(yùn)動(dòng)后不同時(shí)刻取材的組織切片染色顯示肌纖維之間間隙變大,形狀不規(guī)律,呈腫脹狀態(tài);miR-222在運(yùn)動(dòng)后表達(dá)較對(duì)照組整體上調(diào)。一次性長(zhǎng)時(shí)間下坡跑運(yùn)動(dòng)可導(dǎo)致大鼠股直肌損傷,損傷后2周肌纖維細(xì)胞結(jié)構(gòu)基本恢復(fù)正常,通過血清CK的變化特點(diǎn),血清CK可作為運(yùn)動(dòng)性骨骼肌損傷評(píng)價(jià)的敏感指標(biāo)之一。一次性下坡跑誘導(dǎo)的運(yùn)動(dòng)性骨骼肌損傷中miR-222表達(dá)上調(diào),促進(jìn)了骨骼肌損傷的修復(fù)。
[Abstract]:To explore the expression level of miR-222 in skeletal muscle injury and repair, this study selected skeletal muscle injury induced by treadmill, experimental animal model according to Armstrong, a total of 2 month old healthy male SD rats, the establishment of the rectus femoris injury model, drawn at different time after the end of the downhill running, to detect the serum level of CK, the frozen sections of the rectus femoris (HE staining for histological analysis; transverse), using RT-PCR test to detect the level of miR-222. Serum CK in the rectus femoris muscle after exercise injury increased continuously in the process of expression; HE staining: quiet control group showed closely arranged rules, muscle fiber size uniform, polygonal; compared with the control group after exercise, different time from tissue staining showed that the muscle fiber gap between the larger, irregular shape, a swelling state; at miR-222 after exercise as compared with the control group, the overall increase. A long time downhill running can lead to rat rectus femoris injury, 2 week muscle fiber cell structure returned to normal after injury, through the changes of serum CK, serum CK can be used as one of the sensitive indicators of exercise-induced skeletal muscle injury evaluation. The one-time downhill up-regulated miR-222 exercise-induced skeletal muscle injury induced by running, promote the repair of skeletal muscle injury.
