【摘要】：【目的]用大鼠放射性肺損傷模型模擬臨床治療,探討培美曲塞聯(lián)合照射是否會(huì)加重正常肺組織的損傷。 [方法]健康成年雄性Wister大鼠40只,隨機(jī)分為正常對(duì)照組、單純照射組、單純藥物組和藥物照射同步組。照射組采用6MV-X線12Gy單次照射大鼠雙肺尖,藥物組采用培美曲塞20mg/kg單次腹腔注射,正常對(duì)照組予等劑量生理鹽水和假照。分別于實(shí)驗(yàn)前1天、實(shí)驗(yàn)后第7、21、35、49天處死各組大鼠2只,行HE染色比較觀察各組大鼠肺組織病理形態(tài)學(xué)改變、ELISA法測(cè)定血清轉(zhuǎn)化生長(zhǎng)因子β1(TGF-β1)的表達(dá)水平。 [結(jié)果]受過照射的大鼠經(jīng)歷了放射性肺炎到放射性肺纖維化的過程,肺組織逐漸出現(xiàn)炎癥細(xì)胞積聚、肺間質(zhì)充血水腫、肺泡壁增厚、肺組織結(jié)構(gòu)消失和膠原纖維沉積；而藥物組和對(duì)照組大鼠肺組織無明顯異常變化。實(shí)驗(yàn)后第7天,單純照射組和藥物照射同步組大鼠血清TGF-β1的含量開始增高并持續(xù)到第49天,但兩組之間無統(tǒng)計(jì)學(xué)差異(P0.05),單純藥物組和正常對(duì)照組TGF-β1的含量一直維持在正常水平,照射組分別與藥物組、對(duì)照組相比較,未見統(tǒng)計(jì)學(xué)差異(P0.05)。 [結(jié)論]培美曲塞聯(lián)合照射不會(huì)加重正常肺組織的損傷,對(duì)臨床肺腺癌的同步或序貫化放療的應(yīng)用有一定的參考價(jià)值。
[Abstract]:[objective] to study whether the combined irradiation of pemetrexide can aggravate the injury of normal lung tissue by simulating the clinical treatment of rats with radiation-induced lung injury. [methods] Forty healthy adult male Wister rats were randomly divided into normal control group, simple irradiation group, single drug group and drug irradiation synchronous group. The drug group was given a single intraperitoneal injection of Pemetrexed 20mg / kg, and the control group was given the same dose of normal saline and false radiography. Two rats in each group were killed on the first day before the experiment and 3549 days after the 7th day of the experiment. The expression of TGF- 尾 1 in serum of rats with HE staining was measured by Elisa. [results] the irradiated rats experienced the process of radiation pneumonia to radiation-induced pulmonary fibrosis. Inflammatory cell accumulation, interstitial congestion and edema, alveolar wall thickening, disappearance of lung tissue structure and deposition of collagen fibers appeared in the lung tissue. On the 7th day after the experiment, the serum TGF- 尾 1 content of the rats in the single irradiation group and the drug irradiation synchronic group began to increase and continued until the 49th day after the experiment, but there was no obvious abnormal change in the lung tissue of the rats in the drug group and the control group. However, there was no statistical difference between the two groups. The content of TGF- 尾 1 in the drug group and the normal control group was maintained at the normal level, and there was no statistical difference between the irradiation group and the drug group, compared with the control group, there was no statistical difference in the content of TGF- 尾 1 between the two groups. [conclusion] combined pemetrexide irradiation can not aggravate the injury of normal lung tissue, and has certain reference value in the application of synchronous or sequential radiotherapy for clinical lung adenocarcinoma.
