γ射線照射對(duì)小鼠皮膚缺損創(chuàng)面愈合的影響及其機(jī)制探討

【摘要】:目的觀察γ射線照射對(duì)小鼠皮膚缺損創(chuàng)面愈合的影響,并探討其可能的機(jī)制。方法將60只健康雄性C57BL/6小鼠隨機(jī)分為觀察組和對(duì)照組,各30只。觀察組采用60Co-γ射線(6 Gy)照射小鼠全身5 min,照射后腹腔麻醉消毒,于小鼠背部造成深及筋膜的1 cm×1 cm大小皮膚缺損創(chuàng)面,用TegadermTM貼膜覆蓋創(chuàng)面后隔日換藥。對(duì)照組不予射線照射,其余處理同觀察組。兩組于造模后第3天(T0)、7天(T1)、14天(T2),采用Image Proplusv1.5圖像分析系統(tǒng)測(cè)量殘余創(chuàng)面面積,記錄創(chuàng)面組織新生毛細(xì)血管及成纖維細(xì)胞數(shù)量,采用Real-time PCR法檢測(cè)創(chuàng)面組織血管內(nèi)皮生長(zhǎng)因子(VEGF)、堿性成纖維因子2(FGF-2)、血小板衍生因子(PDGF)、轉(zhuǎn)化生長(zhǎng)因子β(TGF-β)mRNA相對(duì)表達(dá)量。結(jié)果隨著時(shí)間的延長(zhǎng),兩組殘余創(chuàng)面面積逐漸減小,創(chuàng)面新生毛細(xì)血管及成纖維細(xì)胞數(shù)量均先升高后降低。觀察組各時(shí)點(diǎn)殘余創(chuàng)面面積均超過(guò)對(duì)照組,T0時(shí)點(diǎn)新生毛細(xì)血管數(shù)量和T0、T1時(shí)點(diǎn)成纖維細(xì)胞數(shù)量均低于對(duì)照組,P均0.05。觀察組T0、T1時(shí)點(diǎn)VEGF、PDGF、TGF-β和各時(shí)點(diǎn)FGF-2 mRNA相對(duì)表達(dá)量均低于對(duì)照組,P均0.05。結(jié)論γ射線照射可以導(dǎo)致小鼠皮膚缺損創(chuàng)面愈合延遲,可能與其降低血管生成因子和成纖維因子表達(dá),從而抑制新生血管和肉芽組織形成有關(guān)。
[Abstract]:Objective to observe the effect of 緯 -ray irradiation on the healing of skin defect wound in mice and to explore its possible mechanism. Methods 60 healthy male C57BL / 6 mice were randomly divided into observation group and control group. 30 mice in each group were treated with 60Co- 緯 -ray irradiation for 5 mins, and then disinfected by intraperitoneal anesthesia, resulting in 1 cm 脳 1 cm deep skin defect on the back of the mice. The wounds were covered with TegadermTM film every other day. The control group was not irradiated by radiation, the other treatment was the same as the observation group. The area of residual wound was measured by Image Proplusv1.5 image analysis system. The number of new capillaries and fibroblasts in wound tissue were recorded. Real-time PCR method was used to detect the relative expression of vascular endothelial growth factor (VEGF), basic fibroblast factor (2FGF-2), platelet-derived factor (PDGF-), transforming growth factor 尾 (TGF- 尾) in wound tissue. The area of residual wound in observation group was higher than that in control group at T _ 0 and the number of fibroblast at T _ 0T _ 1 was lower than that in control group. The relative expression of VEGF PDGF- 尾 and FGF-2 mRNA in observation group was lower than that in control group (P < 0.05). Conclusion 緯 -ray irradiation can cause delayed healing of skin defect wound in mice. It may be related to the inhibition of angiogenesis and granulation tissue formation by reducing the expression of angiogenic factor and fibroblast factor.
