有氧運(yùn)動(dòng)對(duì)MI后大鼠VEGF、bFGF分泌及心臟血管新生的影響研究

【摘要】:研究目的:探討不同時(shí)長(zhǎng)有氧運(yùn)動(dòng)對(duì)心肌梗死(MI)后大鼠血管內(nèi)皮生長(zhǎng)因子(VEGF)、堿性成纖維細(xì)胞生長(zhǎng)因子(bFGF)分泌情況和堿性成纖維細(xì)胞生長(zhǎng)因子受體(FGFR)的mRNA表達(dá)水平,以及對(duì)心臟血管新生的影響。研究方法:選用雄性8周齡SD大鼠40只,隨機(jī)分為假手術(shù)組(Sham)、心肌梗死組(MI)、梗死+運(yùn)動(dòng)2周組(ME2),梗死+運(yùn)動(dòng)8周組(ME8),每組10只。以冠狀動(dòng)脈左前降支(LAD)結(jié)扎術(shù)構(gòu)建MI模型,ME組術(shù)后1周開(kāi)始運(yùn)動(dòng)。運(yùn)動(dòng)方案為:適應(yīng)性運(yùn)動(dòng)1周后,每天10 m/min熱身10 mmin,然后以高強(qiáng)度20 m/min運(yùn)動(dòng)8min,再以低強(qiáng)度15 m/min運(yùn)動(dòng)2 min,高低強(qiáng)度交替4個(gè)循環(huán),最后20 m/min運(yùn)動(dòng)8min,10 m/min運(yùn)動(dòng)2min后結(jié)束。每天訓(xùn)練1次,5 d/w,共訓(xùn)練8w。以二維超聲心動(dòng)圖檢測(cè)大鼠心臟室壁厚度、射血功能;大鼠心肌組織血管新生情況分別采用透射電鏡掃描、Westernblot檢測(cè)心肌組織CD34的表達(dá)水平來(lái)衡量;采用ELISA的方法檢測(cè)大鼠血漿、心肌組織VEGF、 bFGF分泌水平,RT-PCR檢測(cè)FGF-R的mRNA水平。心室研究結(jié)果:(1)心梗使大鼠的心臟重量損失,有氧運(yùn)動(dòng)后心臟重量損失有明顯的改善作用,有氧運(yùn)動(dòng)8周使心重/體重比值(HW/BW)明顯增加(P≤0.05)、左心室重量/體重比值明顯增加(P≤0.05)。(2)心肌梗死手術(shù)使大鼠心臟心室壁變薄,射血分?jǐn)?shù)(EF)顯著降低(P≤0.01);有氧運(yùn)動(dòng)使MI大鼠的室壁厚度增加、射血功能加強(qiáng),其中ME8組舒張期左室后壁厚度(LVPWd)與舒張期室間隔厚度之和明顯高于MI組(P0.05), ME8組EF比MI組明顯升高(P≤0.05)。(3)電鏡觀察結(jié)果顯示,MI使心肌組織血管內(nèi)皮細(xì)胞損傷、間隙增大,基底膜不完整,ME2和ME8組均能見(jiàn)到新生的微小血管。CD34的Westernblot結(jié)果顯示,ME2組的CD34表達(dá)水平比MI組明顯升高(P≤0.05),提示血管新生程度活躍。(4)有氧運(yùn)動(dòng)使血漿和心肌組織的VEGF分泌水平明顯升高,其中ME2組血漿的VEGF水平比MI組明顯升高(P≤0.05),ME8組血漿的VEGF水平比MI組明顯著升高(P≤0.01),且明顯高于ME2組(P≤0.05);ME2組(P≤0.01)和ME8組(P≤0.01)心肌組織的VEGF水平都顯著高M(jìn)I組。(5)有氧運(yùn)動(dòng)使血漿和心肌組織的bFGF分泌水平明顯升高,其中ME2組(P≤0.01)和ME8組(P≤0.01)血漿的bFGF水平比MI組都有顯著升高;ME2組(P≤0.01)和ME8組(P≤0.01)心肌組織的bFGF水平也都顯著高于MI組。(6)有氧運(yùn)動(dòng)使bFGF-R的mRNA表達(dá)水平明顯升高,ME2組(P≤0.05)、ME8組(P≤0.05)與MI組相比,bFGF-R的mRNA水平都有明顯升高。研究結(jié)論:(1)有氧運(yùn)動(dòng)有利于MI大鼠心臟舒縮功能的改善;(2)有氧運(yùn)動(dòng)能促進(jìn)MI大鼠心肌組織血管新生;(3)有氧運(yùn)動(dòng)可能是通過(guò)上調(diào)VEGF、 bFGF的分泌水平,以及bFGF-R的mRNA表達(dá)水平來(lái)促進(jìn)血管新生的。
[Abstract]:Objective: to investigate the effects of aerobic exercise on vascular endothelial growth factor (VEGF) after myocardial infarction (MI) in rats. The secretion of basic fibroblast growth factor (bFGF) and the mRNA expression of basic fibroblast growth factor receptor (BFGFR). Methods: forty male 8-week-old SD rats were randomly divided into sham-operated group and myocardial infarction group (MIM). The MI model was established by ligation of left anterior descending branch of coronary artery (LAD). The ME group began to exercise 1 week after the operation. The exercise plan was: after 1 week of adaptive exercise, 10 m / min, 10 min / min daily, and then 8 minutes of high intensity 20 m / min exercise. Low intensity 15m / min exercise for 2 min, high and low intensity alternating 4 cycles, and last 20 m / min exercise 8 minutes. 10 m / min exercise lasted 2 minutes. The rats were trained once a day for 5 d / w for 8 weeks. The ventricular wall thickness and ejection function were measured by two dimensional echocardiography. The expression of CD34 in myocardial tissue was measured by transmission electron microscope scanning and Western blot. The levels of VEGF and bFGF in plasma and myocardial tissue of rats were measured by ELISA. RT-PCR was used to measure the mRNA level of FGF-R. Ventricular study results showed that myocardial infarction could significantly improve cardiac weight loss after aerobic exercise in rats.
