hUC-MSCs-CM和NAC對(duì)輻射后神經(jīng)元氧化應(yīng)激及增殖與凋亡的影響

目的 探討人臍帶間充質(zhì)干細(xì)胞條件培養(yǎng)基(human umbilical cord mesenchymal stem cells conditioned medium,h UC-MSCs-CM)和N-乙酰半胱氨酸(Nacetylcysteine,NAC)對(duì)輻射相關(guān)氧化應(yīng)激和海馬神經(jīng)元增殖及凋亡的影響。方法 用不同劑量(0、5、10、15Gy)的X射線分別輻射HT-22細(xì)胞,篩選最佳輻射劑量。應(yīng)用酶消化法提取人臍帶間充質(zhì)干細(xì)胞(human umbilical cord mesenchymal stem cells,h UC-MSCs),經(jīng)培養(yǎng)、傳代后于顯微鏡下觀察形態(tài),流式細(xì)胞儀檢測細(xì)胞表面標(biāo)志,選取處于指數(shù)生長期的第3代細(xì)胞,用無血清培養(yǎng)基培養(yǎng)48h,收集上清用于后續(xù)實(shí)驗(yàn)。實(shí)驗(yàn)分組:空白對(duì)照組(Control),單純輻射組(RT),輻射+MSCs-CM組(RT+MSCs-CM),單純MSCs-CM組(MSCsCM),輻射+NAC組(RT+NAC),輻射+MSCs-CM+PI3K抑制劑組(LY294002)。除Control組和MSCs-CM組外,其余各組均給予10Gy X線進(jìn)行輻射。輻射...
