低濃度鎘對人腎系膜細胞和腎足細胞功能的作用及機制研究

研究背景鎘(cadmium,Cd)是一種常見的環(huán)境毒物和工業(yè)污染物,可通過被污染的食物、飲用水、吸煙和職業(yè)接觸等途徑進入人體。鎘的生物半衰期較長并可以蓄積于不同靶器官,鎘暴露會嚴重危害人體健康,它可以對骨骼、腎臟、肝臟、生殖和免疫等器官產(chǎn)生毒性作用,其中腎臟是鎘產(chǎn)生危害的重要靶器官。鎘可以誘導(dǎo)肝臟產(chǎn)生金屬硫蛋白(Metallothionein,MT)并結(jié)合成Cd-MT,Cd-MT隨血液循環(huán)轉(zhuǎn)移至腎臟和其他靶器官。鎘可通過應(yīng)激反應(yīng)損傷腎小球、近曲小管和遠曲小管,在血液循環(huán)中以自由離子或和血漿蛋白結(jié)合以結(jié)合金屬硫蛋白的形式存在,經(jīng)腎小球濾過,可以直接損傷腎小球?qū)е碌鞍啄?鎘蓄積還可以造成腎小管重吸收功能障礙。腎小球細胞主要由腎小球內(nèi)皮細胞、腎系膜細胞和腎足細胞組成,我們的前期研究發(fā)現(xiàn):低濃度鎘對腎小球內(nèi)皮細胞功能無顯著影響,然而低濃度鎘對腎系膜細胞和腎足細胞功能的影響尚不明確,需要進一步研究。c-Jun氨基末端激酶(JNK)家族是促分裂原活化蛋白激酶(MAPKs)成員之一,可被細胞因子、生長因子、應(yīng)激等多種因素激活,參與細胞增殖與分化、細胞形態(tài)維持、細胞骨架構(gòu)建等多種生物學(xué)反應(yīng)。JNK被上...
