雙酚A對(duì)小鼠心臟射血功能及5種蛋白表達(dá)的影響

【摘要】:[目的]探討雙酚A(BPA)對(duì)雄性小鼠心肌射血功能的影響及潛在分子機(jī)制。[方法]以C57/BL6雄性小鼠為實(shí)驗(yàn)動(dòng)物,持續(xù)4周,每天腹腔注射染毒BPA,實(shí)驗(yàn)組染毒濃度分別為1、5、10 mg/kg,對(duì)照組采用同等體積生理鹽水處理。應(yīng)用小鼠超聲儀觀察左心室形態(tài)及功能的變化,采用Western blot技術(shù)檢測(cè)小鼠心室組織相關(guān)的蛋白表達(dá)變化。[結(jié)果]與對(duì)照組相比,BPA 10 mg/kg組小鼠的左心室射血分?jǐn)?shù)(LVEF)[(64.79±6.17)%vs(57.53±3.03)%,P0.05]及左心室短軸縮短分?jǐn)?shù)(LVFS)[(34.46±2.62)%vs(29.01±1.74)%,P0.05]降低,而左心室舒張末期容積(LVEDV)值增高[(32.99±5.63)μL vs(37.48±2.72)μL,P0.05]。BPA染毒組伽馬組蛋白2A變異體(γH2AX)的蛋白表達(dá)水平顯著增加。A型利鈉肽(ANP)的蛋白表達(dá)水平呈現(xiàn)先增高后下降的趨勢(shì)。中、高劑量BPA組小鼠心肌組織細(xì)胞外調(diào)節(jié)蛋白激酶(ERK)及絲氨酸/蘇氨酸激酶1(Akt)的磷酸化蛋白表達(dá)水平顯著增加。通過對(duì)比,各處理組的Brac-1蛋白表達(dá)水平差異無統(tǒng)計(jì)學(xué)意義。[結(jié)論]BPA可引起小鼠心室功能異常,隨著BPA染毒劑量的增加,小鼠的LVEF及LVFS降低,與BPA引起的蛋白激酶pAkt及pERK表達(dá)上調(diào)呈負(fù)相關(guān)。
[Abstract]:Objective to investigate the effect of bisphenol A (BPA) on myocardial ejection function in male mice and its potential molecular mechanism. [methods] C57/BL6 male mice were used as experimental animals for 4 weeks. The concentrations of BPA, in the experimental group were 1 and 5, respectively, and the 10 mg/kg, control group was treated with the same volume of saline. The changes of left ventricular morphology and function were observed by ultrasound in mice, and the expression of related proteins in ventricular tissue of mice was detected by Western blot technique. [results] compared with the control group, the left ventricular ejection fraction (LVEF) [(64.79 鹵6.17)% vs (57.53 鹵3.03)%, P 0.05] and the shortening fraction of left ventricular short axis (LVFS) [(34.46 鹵2.62)% vs (29.01 鹵1.74)%, P 0.05] in BPA 10 mg/kg group decreased, while the (LVEDV) value of left ventricular end diastolic volume increased [(32.99 鹵5.63) 渭 L vs (3748 鹵2.72) 渭 L, but the left ventricular end diastolic volume decreased [(34.46 鹵2.62)% vs (29.01 鹵1.74)%, P 0.05]. The protein expression level of gamma histone 2A variant (gamma H2AX) increased significantly in BPA exposed group. The protein expression level of type A natriuretic peptide (ANP) increased at first and then decreased. The expression of extracellular regulated protein kinase (ERK) and serine / threonine kinase 1 (Akt) phosphorylation protein in high dose BPA group was significantly increased. There was no significant difference in the expression of Brac-1 protein among the treatment groups. [conclusion] BPA can induce abnormal ventricular function in mice. With the increase of BPA dose, LVEF and LVFS in mice decrease, which is negatively correlated with the up-regulation of protein kinase pAkt and pERK expression induced by BPA.
