硫酸鈹致小鼠肺線粒體通透性轉(zhuǎn)換孔高通透性開放及細胞凋亡研究

【摘要】:目的研究腹腔注射硫酸鈹(Be SO4·4H2O)溶液致小鼠肺線粒體通透性轉(zhuǎn)換孔(m PTP)的高通透性開放及細胞凋亡效應。方法 6周齡雄性昆明小鼠36只隨機分成3組,每組12只:空白對照組、1.0 mg/kg Be SO4組和2.0 mg/kg Be SO4組,分別按0.1 ml/10 g(體重)腹腔注射滅菌生理鹽水、1.0、2.0 mg/kg Be SO4滅菌生理鹽水,均隔天注射,一月后處死小鼠取肺臟:差速離心制備線粒體懸液檢測線粒體活性氧(ROS)、跨膜電位(Δψm)和m PTP開放度;提取蛋白測定細胞色素C(Cyt-c)、Bax、Bcl-2的表達;電子顯微鏡觀察肺細胞超微結(jié)構(gòu)。結(jié)果與對照組比較,硫酸鈹染毒組小鼠肺線粒體ROS含量、PTP開放度、細胞色素C和Bax表達均增加(P0.05),Δψm、Bcl-2及Bcl-2/Bax比例均降低(P0.05);染毒組小鼠肺上皮細胞絨毛消失,細胞核固縮,凋亡特征明顯。結(jié)論腹腔注射硫酸鈹可致小鼠肺細胞線粒體通透性轉(zhuǎn)換孔高通透性開放及肺細胞凋亡。
[Abstract]:Objective to study the effects of intraabdominal injection of beryllium sulfate (Be SO4 路4H2O) solution on high permeability opening and apoptosis of (m PTP) in mouse lung mitochondrial permeability transition pore. Methods 36 6-week-old male Kunming mice were randomly divided into 3 groups with 12 mice in each group: blank control group, 1.0 mg/kg Be SO4 group and 2.0 mg/kg Be SO4 group, respectively, treated with 0.1 ml/10 g (body weight) intraabdominal injection of sterilized saline and 1.0 mg/kg Be SO4 sterilized saline, all of which were injected with saline every other day. One month later, the mice were killed and the lungs were taken. The mitochondrial suspension was prepared by differential centrifugation to detect the transmembrane potential (螖 (ROS), m) and m PTP openness of reactive oxygen species (Ros). The expression of cytochrome C (Cyt-c) and Bax,Bcl-2 were measured by extracted protein, and the ultrastructure of lung cells was observed by electron microscope.
