苦蕎提取物成分分析及不同極性提取物對α-淀粉酶的抑制作用

【摘要】:采用石油醚、乙酸乙酯與正丁醇萃取獲得不同極性的苦蕎提取物,采用超高效液相色譜-串聯(lián)四極桿飛行時間質(zhì)譜法分析提取物的成分。結(jié)果表明苦蕎提取物中含有大量的黃酮類物質(zhì),主要包括蘆丁、槲皮素、異槲皮素及其糖苷等,還含有羥基苯甲酸等酚酸類物質(zhì)。再以抑制率、抑制類型、熒光猝滅效應(yīng)實驗分析提取物對α-淀粉酶的抑制作用。結(jié)果表明,苦蕎提取物對α-淀粉酶的抑制作用表現(xiàn)為乙酸乙酯相提取物、正丁醇相提取物與粗提物的結(jié)果類似,都有較為顯著的抑制作用,而石油醚相提取物則沒有明顯且規(guī)律的抑制效果,可推斷出苦蕎提取物中各成分共同發(fā)揮了對α-淀粉酶活性的抑制作用,且抑制類型為非競爭性抑制。通過熒光猝滅實驗得知抑制的機理為苦蕎提取物中的成分與α-淀粉酶結(jié)合,從而使α-淀粉酶的熒光特性發(fā)生靜態(tài)猝滅。
[Abstract]:The extracts of Tartary buckwheat with different polarity were extracted by petroleum ether, ethyl acetate and n-butanol. The constituents of extracts were analyzed by ultra-high performance liquid chromatography-tandem quadrupole time-of-flight mass spectrometry. The results showed that the extract of Tartary buckwheat contained a large number of flavonoids, including rutin, quercetin, isoquercetin and its glycosides, as well as phenolic acids such as hydroxybenzoic acid. Then the inhibitory effect of extract on 偽-amylase was analyzed by inhibition rate, type of inhibition and fluorescence quenching effect. The results showed that the inhibitory effect of Tartary buckwheat extract on 偽-amylase was ethyl acetate extract, and the results of n-butanol phase extract were similar to that of crude extract, all had significant inhibitory effect on 偽-amylase, and the inhibitory effect of n-butanol phase extract was similar to that of crude extract. But the petroleum ether phase extract had no obvious and regular inhibitory effect. It can be concluded that the components of Tartary buckwheat extract play a common role in inhibiting the activity of 偽-amylase, and the type of inhibition is non-competitive inhibition. The mechanism of inhibition was that the components of Tartary buckwheat extract combined with 偽-amylase, thus the fluorescence characteristics of 偽-amylase were quenched statically.
