人血中有機鹵代化合物的水平及其與甲狀腺指標的相關(guān)性研究

【摘要】:持久性有機物污染物(Persistent Organic Pollutants, POPs)所引起的環(huán)境問題日益嚴重,對人類和生態(tài)環(huán)境都具有很大的潛在威脅,受到各國各界的重視。目前十溴聯(lián)苯醚在人血中的含量正逐年增高,而且其在環(huán)境中及人體中還可能發(fā)生代謝或降解作用,生成低溴代的或羥基化的PBDE,其對人體健康的影響不容忽視。而PCB和OCPs雖然已禁止生產(chǎn)和使用,但由于其持久性、難降解、脂溶性及生物富集作用,其在人體中仍將存在一段時間。新型阻燃劑DP,其使用其實已有四十多年的歷史,但是近幾年才開始引起人們的注意,人血中DP水平數(shù)據(jù)及毒理學(xué)數(shù)據(jù)缺乏,健康影響不明確。 本文建立了凝膠滲透色譜(GPC)去脂、氣相色譜-負化學(xué)電離源-質(zhì)譜(GC-NCI-MS)法測定PBDEs、PCBs、DP及OCPs的方法。研究采集并測定了濰坊及南寧各40個人血清樣本中上述化合物濃度。本研究發(fā)現(xiàn)濰坊地區(qū)Σ4PBDEs (BDE-100、99、153及183)濃度范圍在3.4-62ng/g脂重,平均濃度為11ng/g脂重,中值為6.4ng/g脂重,BDE-209范圍從ND-100000ng/g脂重,均值和中值分別為1200和440ng/g脂重。而Σ2PCBs (BDE-180、170)濃度范圍在ND-14ng/g脂重,平均濃度為3.2ng/g脂重,中值為2.5ng/g脂重!艱P濃度范圍在ND-36ng/g脂重,平均濃度為3.6ng/g脂重,中值為1.8ng/g脂重,fanti值從0.27到0.93不等,均值為0.65。HCB及ΣDDT濃度范圍分別為57-420ng/g脂重和49-38000ng/g脂重,平均濃度分別為130ng/g脂重和3100ng/g脂重,中值分別為110ng/g脂重和1200ng/g脂重。 而南寧地區(qū)Σ4PBDEs在2.0-14ng/g脂重,平均濃度為5.0ng/g脂重,中值為4.80ng/g脂重。BDE-209范圍從ND-6300ng/g脂重,均值和中值分別為720和270ng/g脂重。∑2PCB在ND-4.9ng/g脂重,平均濃度為1.4ng/g脂重,中值為1.3ng/g脂重。ΣDP濃度范圍在ND-20ng/g脂重,平均濃度為3.3ng/g脂重,中值為1.8ng/g脂重,fanti值從0.35到0.95不等,均值為0.71。HCB及ΣDDT濃度范圍為26-240ng/g脂重和ND-11000ng/g脂重,平均濃度分別為63ng/g脂重和1400ng/g脂重,中值分別為53ng/g脂重和760ng/g脂重。 本研究中檢出的PBDEs. PCBs及OCPs均顯示出濰坊高于南寧的趨勢,統(tǒng)計學(xué)結(jié)果也顯示兩地BDE-100、99、153,PCB-180、170及HCB的水平存在顯著性差異。而兩地DP水平相當,統(tǒng)計學(xué)上也無顯著性差異,而且兩地fanti值較高,說明兩地DP組成以anti-DP為主。 本研究同時對濰坊及南寧人血清樣品5項甲狀腺指標做了檢測,80個樣本總異常率達到了50%。對4類有機鹵代化合物與甲狀腺功能指標的相關(guān)關(guān)系,多元線性回歸分析結(jié)果表明山東人群fT3水平可能受BDE-183及p-HCH的影響,而且前者占主導(dǎo)影響;而南寧人群fT3水平則可能受PCB-180及PCB-170的影響,并以PCB-170為主。雖然本研究中統(tǒng)計學(xué)并未發(fā)現(xiàn)更多4類有機鹵代化合物與甲狀腺功能指標的相關(guān)性,但是兩地甲狀腺指標異常率之高不容忽視,更多研究包括動物毒性實驗應(yīng)該深入進行,以評價其健康影響。
[Abstract]:The environmental problems caused by persistent organic pollutants (Persistent Organic Pollutants, POPs) are becoming more and more serious, which have a great potential threat to human and ecological environment. At present, the content of decabromodiphenyl ether in human blood is increasing year by year, and it may be metabolized or degraded in the environment and human body. The effect of decabromodiphenyl ether on human health can not be ignored. Although PCB and OCPs have been banned from production and use, they will still exist in human body for some time due to their persistence, refractory degradation, liposolubility and bioconcentration. The new flame retardant DP, has been used for more than 40 years, but in recent years, it has attracted people's attention. The data of DP level and toxicology in human blood are lacking, and the health effects are not clear. A method for the determination of PBDEs,PCBs,DP and OCPs by gel permeation chromatography (GPC) and gas chromatography-negative chemical ionization source-mass spectrometry (GC-NCI-MS) was established. The concentrations of these compounds in 40 serum samples from Weifang and Nanning were determined. In this study, the concentrations of 危 4PBDEs (BDE-100,99153 and 183) in Weifang area were found to be in the range of 3.4-62ng/g lipid weight, the average concentration was 11ng/g lipid weight, the median was 6.4ng/g lipid weight, and the BDE-209 range was ND-100000ng/g lipid weight.
