魚油對(duì)PM2.5所致心血管炎性和氧化損傷拮抗作用

【摘要】:目的探討深海魚油對(duì)大氣細(xì)顆粒物(PM2.5)引起的心血管系統(tǒng)中的炎性損傷和氧化應(yīng)激損傷影響。方法將SD雄性大鼠隨機(jī)分為魚油低、中、高劑量組,魚油對(duì)照組,溶劑對(duì)照組(玉米油)和PM2.5染毒組;魚油組大鼠先進(jìn)行深海魚油灌胃28 d,再進(jìn)行PM2.5氣管滴注染毒隔天1次,共3次,PM2.5染毒期間同時(shí)進(jìn)行魚油灌胃,PM2.5染毒組先正常飼養(yǎng)28 d,再進(jìn)行PM2.5染毒;取血清檢測(cè)白細(xì)胞介素-6(IL-6)、腫瘤壞死因子α(TNF-α)、超敏C反應(yīng)蛋白(hs-CRP)、丙二醛(MDA)、總超氧化物歧化酶(T-SOD)、谷胱甘肽(GSH)和谷胱甘肽過(guò)氧化物酶(GSH-Px)。結(jié)果與溶劑對(duì)照組比較,PM2.5染毒組大鼠血清中IL-6、TNF-α、hs-CRP、MDA含量[分別為(24.94±2.06)、(821.45±14.26)ng/m L、(3.10±0.28)mg/L、(15.88±1.41)nmol/m L]升高,GSH、GSH-Px、TSOD活性[(4.62±0.37)μmol/L、(289.28±30.65)活力單位、(239.26±4.97)U/m L]降低(均P0.05);與PM2.5染毒組比較,魚油高劑量組大鼠血清中IL-6、TNF-α、hs-CRP、MDA含量[分別為(16.67±0.23)、(73.01±3.80)ng/m L、(1.36±0.22)mg/L、(11.82±0.79)nmol/m L]下降,GSH、GSH-Px、T-SOD活性[(24.05±3.83)μmol/L、(531.33±40.39)活力單位、(273.36±3.92)U/m L]升高,呈劑量效應(yīng)關(guān)系(均P0.05)。結(jié)論深海魚油對(duì)大氣PM2.5所致心血管系統(tǒng)炎性反應(yīng)和氧化應(yīng)激損傷具有明顯拮抗作用。
[Abstract]:Objective to investigate the effects of deep-sea fish oil on inflammatory injury and oxidative stress injury in cardiovascular system induced by atmospheric fine particulate matter (PM2.5). Methods male SD rats were randomly divided into low, medium and high dose groups, fish oil control group, solvent control group (corn oil) and PM2.5 exposure group. The rats in the fish oil group were fed with deep-sea fish oil for 28 days and then injected with PM2.5 once every other day for 3 times. The rats in the PM2.5 group were fed with fish oil at the same time during PM2.5 exposure. The rats in the PM2.5 group were fed normally for 28 days and then PM2.5. Serum levels of interleukin-6 (IL-6), tumor necrosis factor 偽 (TNF- 偽), hypersensitive C-reactive protein (hs-CRP) and malondialdehyde (MDA),) total superoxide dismutase (T-SOD) were measured. Glutathione (GSH) and glutathione peroxidase (GSH-Px). Results compared with solvent control group, the contents of IL-6,TNF- 偽 and hs-CRP,MDA in serum of rats exposed to PM2.5 were (24.94 鹵2.06), (821.45 鹵14.26) ng/m L, (3.10 鹵0.28) mg/L, respectively. The activity of GSH,GSH-Px,TSOD [(4.62 鹵0.37) 渭 mol/L, (289.28 鹵30.65), (239.26 鹵4.97) U / mL] was increased (P 0.05). Compared with PM2.5 exposure group, the contents of IL-6,TNF- 偽 and hs-CRP,MDA in serum of high dose group of fish oil were (16.67 鹵0.23), (73.01 鹵3.80) ng/m L, (1.36 鹵0.22) mg/L, respectively. The activity of GSH,GSH-Px,T-SOD [(24.05 鹵3.83) 渭 mol/L, (531.33 鹵40.39), (273.36 鹵3.92) U / mL] decreased, and the activity of GSH,GSH-Px,T-SOD increased. There was a dose-effect relationship (P0.05). Conclusion Deep-sea fish oil has obvious antagonistic effect on cardiovascular system inflammatory response and oxidative stress injury induced by atmospheric PM2.5.
