酸性鞘磷脂酶在鎘性腎損害中的變化

【摘要】:目的 觀察酸性鞘磷脂酶(ASMase)在鎘性腎損害過(guò)程中的活性變化及規(guī)律,探討ASMase活性變化與鎘性腎損害的關(guān)系。方法 60只SD雄性大鼠隨機(jī)分為對(duì)照組和實(shí)驗(yàn)組,實(shí)驗(yàn)組動(dòng)物腹腔注射1mg/kg Cd2+,每天1次、每周5次,對(duì)照組注射等體積的生理鹽水。染毒第0、1、3、5和7周末實(shí)驗(yàn)組、對(duì)照組每組各隨機(jī)處死6只動(dòng)物,收集血、尿及腎組織。ELISA法測(cè)尿β2微球蛋白的含量,高效液相色譜法測(cè)血、尿及腎組織中ASMase活性,制作病理切片觀察腎組織病變情況。結(jié)果 隨染毒時(shí)間的延長(zhǎng),腎組織有不同程度的病理改變。與對(duì)照組相比,實(shí)驗(yàn)組動(dòng)物腎臟系數(shù)隨染毒時(shí)間的延長(zhǎng)而增加(P0.05);尿β2微球蛋白含量隨染毒時(shí)間的延長(zhǎng)而增加(P0.05);血、尿、腎組織勻漿上清液中ASMase活性從染毒第3周開(kāi)始隨染毒時(shí)間的延長(zhǎng)而增加(P0.05)。結(jié)論 鎘性腎損害中ASMase活性隨染毒時(shí)間的延長(zhǎng)而增加。
[Abstract]:Objective to observe the activity and regularity of acid sphingolipase (ASMase) in the process of cadmium induced renal injury, and to explore the relationship between the activity of ASMase and cadmium induced renal damage. Methods Sixty Sprague-Dawley male rats were randomly divided into control group and experimental group. 1mg/kg Cd2 was injected intraperitoneally in the experimental group, once a day, 5 times a week, and the control group was injected with saline of the same volume. At the end of the 5th and 7th week of exposure, 6 animals were randomly killed in each group in the control group. The contents of 尾 2 microglobulin in urine, urine and renal tissue were collected by Elisa, and the activity of ASMase in blood, urine and renal tissue were measured by high performance liquid chromatography (HPLC). Pathological sections were made to observe the pathological changes of renal tissue. Results with the prolongation of exposure time, renal tissue had different pathological changes. Compared with the control group, the kidney coefficient of the experimental group increased with the time of exposure (P0.05); the content of urinary 尾 2 microglobulin increased with the prolongation of the exposure time (P0.05); the blood, urine, blood, urine, The activity of ASMase in the supernatant of renal homogenate increased with the prolongation of exposure time from the 3rd week of exposure (P0.05). Conclusion the activity of ASMase in cadmium induced renal injury increased with the prolongation of exposure time.
