蛋白質(zhì)二硫鍵異構(gòu)酶在染砷家兔皮膚中的表達(dá)變化

【摘要】:目的觀察亞砷酸鈉(i As3+)染毒對(duì)家兔皮膚蛋白質(zhì)二硫鍵異構(gòu)酶(protein disulfide isomerase,PDI)的影響,探討PDI與砷致皮膚損害的關(guān)系。方法將30只健康雄性家兔隨機(jī)平均分到對(duì)照組(C)及低(L,0.13mg/千克體重(kg.w))、中(M,0.26mg/kg.w)、中高(MH,0.65mg/kg.w)、高(H,1.30mg/kg.w)劑量染毒組。亞砷酸鈉飲水染毒3個(gè)月后處死,取背部相同位置全厚皮膚樣。雙向凝膠電泳分離樣品中蛋白,Bio-Rad圖像系統(tǒng)分析膠圖,篩選差異表達(dá)蛋白,膠內(nèi)酶切后采用基質(zhì)輔助激光解析電離飛行時(shí)間質(zhì)譜進(jìn)行鑒定。通過實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)(real-time polymerase chain reaction,RT-PCR)檢測樣品中PDI mRNA的表達(dá)。結(jié)果染毒3個(gè)月后,家兔無死亡,各組家兔體重?zé)o明顯變化,M、MH、H組可觀察到家兔背腹部毛發(fā)無光澤、粗糙,H組尤為明顯。皮膚中PDI在M、MH組表達(dá)量下降,在H組表達(dá)量上升。PDI mRNA在MH和H組上調(diào)(F=225.83,P0.001)。結(jié)論亞砷酸鈉染毒后,隨染毒劑量升高,家兔皮膚PDI蛋白表達(dá)量先降低后升高,PDI mRNA逐漸增高,同時(shí)可觀察到家兔毛發(fā)粗糙,提示PDI可能在砷致皮膚損傷中發(fā)揮作用。
[Abstract]:Objective to observe the effect of sodium arsenite (I As3) on (protein disulfide isomeraseisomerase (protein disulfide isomerase) in rabbit skin, and to explore the relationship between PDI and arsenic induced skin damage. Methods 30 healthy male rabbits were randomly divided into three groups: control group (C) 0.13 mg / kg), low (kg.w), 0.26 mg / kg 路w), moderate high (MHN 0.65 mg / kg 路w) and high dose (1.30 mg / kg 路w) groups. Sodium arsenite was killed after drinking water for 3 months. Bio-Rad image system was used to isolate proteins from samples by two-dimensional gel electrophoresis. Differential expression proteins were screened and identified by matrix assisted laser desorption ionization time of flight mass spectrometry (TOF-MS). The expression of PDI mRNA was detected by real-time real-time polymerase chain reaction-polymerase chain reaction (RT-PCR). Results after 3 months of exposure, there was no death in rabbits, and there was no obvious change in body weight in each group. The expression of PDI in the skin decreased in MH group, and increased in H group. PDI mRNA was up-regulated in MH and H group (FF225.83, P0.001). Conclusion after sodium arsenite exposure, the expression of PDI protein in rabbit skin decreases first and then increases gradually with the increase of the dose of arsenite. It is also observed that the hair of rabbits is rough, suggesting that PDI may play a role in arsenic induced skin injury.
