鋁對斑馬魚幼魚的神經(jīng)毒性作用及其轉(zhuǎn)錄組學(xué)研究

【摘要】:目的探討鋁對斑馬魚的神經(jīng)毒性作用及其在轉(zhuǎn)錄水平的機(jī)制。方法選取受精1 d后發(fā)育良好的斑馬魚胚胎,隨機(jī)分為2組,分別給予單純培養(yǎng)基(對照組)和50μg/L濃度的AlCl_3處理6 d(AlCl_3處理組)。檢測幼魚自主運(yùn)動(dòng)行為、活性氧(ROS)釋放及轉(zhuǎn)錄組學(xué)變化。結(jié)果與空白對照組相比,經(jīng)AlCl_3處理后的斑馬魚,表現(xiàn)出運(yùn)動(dòng)軌跡明顯無規(guī)律,游泳速度減慢,差異有統(tǒng)計(jì)學(xué)意義(P0.05);AlCl_3增加斑馬魚幼魚的活性氧釋放(P0.05);轉(zhuǎn)錄組學(xué)數(shù)據(jù)顯示,AlCl_3處理組與對照組相比,Neurogenin 1等19個(gè)與退行性神經(jīng)疾病相關(guān)的基因表達(dá)發(fā)生顯著變化(P0.0001),22個(gè)顯著變化的基因富集在MAPK信號通路。結(jié)論鋁對斑馬魚具有神經(jīng)毒性作用,其分子機(jī)制可能與Neurogenin 1等19個(gè)基因表達(dá)的變化及MAPK信號通路的調(diào)控有關(guān)。
[Abstract]:Objective to investigate the neurotoxicity of aluminum to zebrafish and its mechanism at transcription level. Methods zebrafish embryos were randomly divided into two groups: single medium (control group) and ALCL _ 3 treated with 50 渭 g / L AlCl3 for 6 days (ALCL _ 3 treatment group). The spontaneous exercise behavior, the release of reactive oxygen species (Ros) and transcriptome changes in juvenile were detected. Results compared with the blank control group, the zebrafish treated with AlCl3 showed obvious irregular track and slow swimming speed. The difference was statistically significant (P0.05) that AlCl3 increased the release of reactive oxygen species (P0.05), and transcriptional data showed that the expression of 19 genes related to neurogenin 1 and neurodegenerative neuropathy in the AlCl3 treatment group was significantly different from that in the control group (P0.0001), and 22 were significant. The altered genes are enriched in the MAPK signaling pathway. Conclusion Aluminum has neurotoxic effect on zebrafish and its molecular mechanism may be related to the changes of neurogenin 1 gene expression and the regulation of MAPK signaling pathway.
