新疆庫(kù)車(chē)小白杏杏仁油調(diào)節(jié)大鼠血脂作用及機(jī)制的實(shí)驗(yàn)研究

【摘要】:目的:以新疆庫(kù)車(chē)小白杏杏仁油為研究對(duì)象,對(duì)小白杏杏仁油的營(yíng)養(yǎng)成分進(jìn)行測(cè)定并探討小白杏杏仁油調(diào)節(jié)大鼠血脂作用及其機(jī)制。方法:依據(jù)各營(yíng)養(yǎng)成分測(cè)定國(guó)家標(biāo)準(zhǔn)對(duì)新疆庫(kù)車(chē)小白杏杏仁油的營(yíng)養(yǎng)成分進(jìn)行測(cè)定與評(píng)價(jià)。將50只Wistar雄性大鼠在實(shí)驗(yàn)環(huán)境下經(jīng)基礎(chǔ)飼料適應(yīng)性喂養(yǎng)一周,測(cè)定大鼠血清總膽固醇(TC),甘油三酯(TG),高密度脂蛋白膽固醇(HDL-C)和低密度脂蛋白膽固醇(LDL-C)水平。根據(jù)TC水平隨機(jī)將大鼠分為五組:高劑量組、中劑量組、低劑量組、正常對(duì)照組和模型組。三個(gè)劑量組在給予高脂飼料的同時(shí)灌胃不同劑量(低劑量1ml/kg·d,中劑量2ml/kg·d,高劑量4ml/kg·d)的杏仁油,每日一次,正常對(duì)照組喂以普通飼料,模型組喂以高脂飼料,對(duì)照組和模型組每天給予4ml/kg·d純凈水。八周后抽取大鼠血液離心后測(cè)定血清中的總膽固醇(TC),甘油三酯(TG),高密度脂蛋白膽固醇(HDL-C)和低密度脂蛋白膽固醇(LDL-C)水平;載脂蛋白AI、載脂蛋白B;三碘甲狀腺原氨酸(T3)及胰島素水平;取大鼠肝臟,稱重記錄并制作組織切片并凍存肝臟用實(shí)時(shí)熒光定量法測(cè)定ApoAI和ApoB基因表達(dá)水平。結(jié)果:1.每100g新疆庫(kù)車(chē)小白杏杏仁油中含粗脂肪99.5g、碳水化合物0.2g、水分0.1g、雜質(zhì)0.2g、維生素A0.003mg、ɑ-維生素E0.06mg。2.小白杏杏仁油脂肪酸中含有71%油酸、22.7%亞油酸。3.小白杏杏仁油中含有鈣0.663mg/100g、鈉0.828mg/100g、鉀0.39mg/100g、鐵0.083mg/100g、鋅0.054mg/100g。4.正常對(duì)照組的大鼠肝濕重顯著低于其他各組,高劑量組大鼠的肝濕重低于模型組。5.中、高劑量杏仁油組大鼠的血清TC、TG含量不同程度低于模型組,中、高劑量杏仁油組大鼠血清HDL-C含量顯著高于模型組。6.各劑量組大鼠的ApoAI含量明顯高于模型組;模型組大鼠ApoB含量高于空白組,各劑量組與模型組ApoB含量相比沒(méi)有差別。7.各劑量組和空白組大鼠體內(nèi)胰島素、T3含量顯著高于模型組。8.模型組ApoAI基因表達(dá)水平低于空白對(duì)照組,高劑量組ApoAI基因表達(dá)水平高于模型組。模型組ApoB基因表達(dá)水平高于空白對(duì)照組。結(jié)論:新疆庫(kù)車(chē)小白杏杏仁油中碳水化合物和礦物質(zhì)元素含量豐富,中、高劑量組能降低高血脂大鼠TC、TG水平,提高血清中HDL-C的含量,通過(guò)對(duì)ApoAI、ApoB、胰島素和T3的含量的調(diào)節(jié),增強(qiáng)ApoAI基因表達(dá),,直接或間接影響血脂水平。
[Abstract]:Objective: to determine the nutritional components of small white apricot almond oil in Xinjiang Kuqa apricot almond oil and to explore the effect and mechanism of small white apricot almond oil on regulating blood lipids in rats. Methods: the nutritional components of apricot oil from Kuqa Xiaobai apricot in Xinjiang were determined and evaluated according to the national standards. Fifty male Wistar rats were fed with basic diet for one week to determine the levels of serum total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C). According to TC level, rats were randomly divided into five groups: high dose group, middle dose group, low dose group, normal control group and model group. Almond oil of different doses (low dose 1ml/kg d, middle dose 2ml/kg d, high dose 4ml/kg d) was given to the three dose groups once a day. The normal control group was fed with normal diet and the model group with high fat diet. The control group and the model group were given 4ml/kg d purified water daily. The serum levels of total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) were measured after 8 weeks of blood centrifugation. The levels of triiodothyronine (T3) and insulin were measured by real-time fluorescence quantitative method, and the expression levels of ApoAI and ApoB genes were measured by real-time fluorescence quantitative method. Results: the content of crude fat, carbohydrate, moisture, impurity, vitamin A and vitamin E were 99.5g, 0.2g, 0.2g, 0.003mg, 0.06mg.2.The fatty acids of almond oil contained 71% oleic acid, 22.7wt% linoleic acid .3.The contents of crude fat were 99.5g, 0.2g, 0.2g, 0.2g, 0.003mg, 0.06mg.2.The fatty acids of almond oil contained 71% oleic acid and 22.7wt% linoleic acid. Oil contains calcium 0.663mg / 100g, sodium 0.828mg / 100g, potassium 0.39mg / 100g, iron 0.083mg / 100g, zinc 0.054mg / 100g.4. the wet weight of liver in the normal control group is significantly lower than that in the other groups. The liver wet weight of rats in high dose group was lower than that in model group .5.The content of serum TCN TG in high dose almond oil group was lower than that in model group. The content of serum HDL-C in high dose almond oil group was significantly higher than that in model group. 6. The ApoAI content in each dose group was significantly higher than that in model group, and the content of ApoB in model group was higher than that in blank group. There was no difference in ApoB content between each dose group and model group. The content of insulin T 3 in each dose group and blank group was significantly higher than that in model group. The expression level of ApoAI gene in the model group was lower than that in the blank control group, and the expression level of ApoAI gene in the high dose group was higher than that in the model group. The expression level of ApoB gene in model group was higher than that in blank control group. Conclusion: the content of carbohydrate and mineral elements in the almond oil of Kuqa Xiaobai apricot in Xinjiang is rich. The high dose group can decrease the level of TCU TG and increase the content of HDL-C in serum of rats with hyperlipidemia, and regulate the contents of ApoAIN ApoB, insulin and T3. Enhance the expression of ApoAI gene, directly or indirectly affect the level of blood lipids.
