鉬致小鼠亞急性肝臟毒性研究

【摘要】:目的探討鉬對(duì)小鼠肝臟的亞急性毒性作用。方法7周齡雄性昆明小鼠50只,適應(yīng)性喂養(yǎng)一周,按體重隨機(jī)分為五組,分別為對(duì)照組(生理鹽水)、3.440mg/kg(1/100 LD50)、6.880mg/kg(1/50 LD50)、34.400mg/kg(1/10 LD50)、68.800mg/kg(1/5 LD50)鉬酸鈉染毒組,每組10只。腹腔注射鉬酸鈉進(jìn)行染毒,染毒劑量為0.2ml/10g,每日一次,連續(xù)染毒30天。染毒結(jié)束后,測定小鼠肝臟濕量和臟器系數(shù);肝臟中鉬含量;血清中天門冬氨酸氨基轉(zhuǎn)移酶(AST)、丙氨酸氨基轉(zhuǎn)移酶(ALT)、乳酸脫氫酶(LDH)的含量及肝臟勻漿里的谷胱甘肽過氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)、丙二醛(MDA)的含量;并在光鏡下觀察小鼠肝臟組織病理學(xué)變化。結(jié)果1、染毒結(jié)束后,各組小鼠體重均增加,34.400mg/kg、68.800mg/kg染毒組小鼠體重同對(duì)照組相比差異具有統(tǒng)計(jì)學(xué)意義。2、與對(duì)照組相比,各組小鼠肝臟鉬含量升高,且34.400mg/kg,68.800mg/kg組差異具有統(tǒng)計(jì)學(xué)意義。3、僅68.800 mg/kg鉬酸鈉染毒組小鼠血清中AST和LDH含量明顯高于對(duì)照組,且差異有統(tǒng)計(jì)學(xué)意義。34.400mg/kg和68.800mg/kg鉬酸鈉染毒組小鼠血清ALT含量和對(duì)照組小鼠比較差異具有統(tǒng)計(jì)學(xué)學(xué)意義。4、各組小鼠肝臟勻漿中SOD活力均下降,且6.880mg/kg、34.400mg/kg、68.800mg/kg同對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義。各小組小鼠肝臟勻漿中的GSH-PX活力也隨之降低,34.400mg/kg和68.800mg/kg同對(duì)照組相比差異有統(tǒng)計(jì)學(xué)意義。肝臟勻漿中的MDA含量則隨著染毒劑量的增加而增加,同樣34.400mg/kg和68.800mg/kg染毒組和對(duì)照組相比差異具有統(tǒng)計(jì)學(xué)意義。5、組織病理觀察發(fā)現(xiàn)68.800 mg/kg鉬酸鈉染毒組肝細(xì)胞結(jié)構(gòu)不完整,界限不清晰,形態(tài)大小不一,肝細(xì)胞空泡變性,炎細(xì)胞浸潤,肝索排列紊亂,點(diǎn)狀壞死,中央靜脈狹窄。結(jié)論鉬在小鼠肝臟中有一定的蓄積作用,高劑量的鉬對(duì)小鼠肝臟具有損傷作用。
[Abstract]:Objective to study the subacute toxicity of molybdenum on mouse liver. Methods Fifty seven week-old male Kunming mice were fed adaptively for one week. They were randomly divided into five groups according to their body weight: the control group (normal saline 3.440 mg / kg / 100 LD50 / kg 1.880 mg / kg 1 / 50 LD50 + 34.400 mg / kg / 10 LD50 68.800 mg / kg / L / 5 LD50) sodium molybdate exposure group (10 rats in each group). Sodium molybdate was injected intraperitoneally at a dose of 0.2 ml / 10 g once a day for 30 days. At the end of the exposure, the wet amount of liver and organ coefficient, the content of molybdenum in liver and the content of molybdenum in liver of mice were measured. The contents of aspartate aminotransferase (AST), alanine aminotransferase (alt), lactate dehydrogenase (LDH) and glutathione peroxidase (GSH-PX), superoxide dismutase (SOD), malondialdehyde (MDA) in liver homogenate; The histopathological changes of mouse liver were observed under light microscope. Results 1. The body weight of each group increased by 34.400 mg 路kg ~ (-1) 路kg ~ (-1) 68.800 mg 路kg ~ (-1) 路kg ~ (-1) compared with that of the control group (P < 0.01). The molybdenum content in liver of each group was higher than that of the control group. There was significant difference in serum AST and LDH levels in mice exposed to sodium molybdate for 68.800 mg/kg, and the difference was significant in the group of 68.800 mg / kg of 34.400 mg / kg of molybdate. The serum alt content of mice exposed to 68.800mg/kg and sodium molybdate was significantly higher than that of control group. The activity of sod in liver homogenate of each group was decreased, and there was a significant difference between 6.880 mg / kg and 34.400 mg 路kg ~ (-1) 路kg ~ (-1) and 68.800 mg / kg compared with the control group. The activity of GSH-PX in liver homogenate of each group mice also decreased 34.400 mg / kg and 68.800mg/kg significantly compared with the control group. The content of malondialdehyde (MDA) in liver homogenate increased with the increase of dose. Similarly, the difference between 34.400mg/kg and 68.800mg/kg groups was statistically significant. Histopathological observation showed that the hepatocyte structure of 68.800 mg/kg sodium molybdate group was incomplete. The boundary is not clear, the shape is different, the hepatocyte vacuole degeneration, the inflammatory cell infiltration, the hepatic cord arrangement disorder, the dot-like necrosis, the central vein narrow. Conclusion Molybdenum has a certain accumulative effect in mouse liver, and high dose of Mo can damage the liver of mice.
