ROS介導(dǎo)的氧化應(yīng)激在INH誘導(dǎo)的細(xì)胞毒性中的作用及槲皮素的干預(yù)

【摘要】:為探討ROS介導(dǎo)的氧化應(yīng)激在異煙肼(INH)誘導(dǎo)L-02細(xì)胞毒性中的作用及槲皮素的干預(yù)作用,建立體外培養(yǎng)INH誘導(dǎo)L-02細(xì)胞氧化損傷模型,實(shí)驗(yàn)分為對(duì)照組(A)、INH組(B)、槲皮素低劑量組(C)及槲皮素高劑量組(D)。采用生化分析法檢測(cè)L-02細(xì)胞培養(yǎng)液中天冬氨酸氨基轉(zhuǎn)移酶(AST)和丙氨酸氨基轉(zhuǎn)移酶(ALT)的活性;利用熒光探針檢測(cè)L-02細(xì)胞線粒體內(nèi)活性氧(ROS)水平;應(yīng)用比色法檢測(cè)L-02細(xì)胞內(nèi)丙二醛(MDA)、谷胱甘肽(GSH)的含量以及主要抗氧化物酶的活性。結(jié)果表明,與對(duì)照組相比,INH能顯著增加L-02細(xì)胞培養(yǎng)液中AST和ALT的活性、細(xì)胞線粒體內(nèi)ROS水平及細(xì)胞內(nèi)MDA的含量(P0.01),并顯著減少L-02細(xì)胞內(nèi)GSH的含量及超氧化物歧化酶(SOD)、谷胱甘肽過(guò)氧化物酶(GSH-Px)的活性(P0.01)。與INH組比較,槲皮素低劑量組L-02細(xì)胞培養(yǎng)液中AST的活性、線粒體內(nèi)ROS水平及細(xì)胞內(nèi)MDA的含量明顯降低(P0.05),而細(xì)胞內(nèi)SOD的活性明顯增加(P0.05);高劑量槲皮素能顯著降低L-02細(xì)胞培養(yǎng)液中AST和ALT的活性、細(xì)胞線粒體內(nèi)ROS水平及細(xì)胞內(nèi)MDA的含量(P0.01),并能顯著增高L-02細(xì)胞內(nèi)GSH的含量和主要抗氧化物酶的活性(P0.01)。與槲皮素低劑量組相比,槲皮素高劑量組的保護(hù)效應(yīng)更明顯(P0.05)?梢(jiàn),ROS介導(dǎo)的氧化應(yīng)激在INH誘導(dǎo)的L-02細(xì)胞毒性中發(fā)揮了重要作用,且槲皮素對(duì)INH誘導(dǎo)的L-02細(xì)胞氧化損傷具有保護(hù)作用。
[Abstract]:To investigate the role of Ros mediated oxidative stress in the cytotoxicity of L-02 cells induced by isoniazid (INH) and the intervention of quercetin, a model of oxidative damage of L-02 cells induced by INH in vitro was established. The experiment was divided into two groups: the control group, the low dose group, and the high dose group, the control group, the high dose group and the high dose group, respectively. The activities of aspartate aminotransferase (AST) and alanine aminotransferase (alt) in L-02 cell culture medium were detected by biochemical analysis, and the levels of reactive oxygen species (Ros) in mitochondria of L-02 cells were detected by fluorescence probe. The content of malondialdehyde (MDA) and glutathione (GSH) and the activities of main antioxidant enzymes in L-02 cells were detected by colorimetry. The results showed that the activities of AST and alt in L-02 cell culture medium were significantly increased by Inh compared with the control group. The contents of Ros and MDA in mitochondria were significantly decreased in L-02 cells, and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in L-02 cells were significantly decreased (P 0.01). Compared with INH group, the AST activity in L-02 cell culture medium of quercetin low-dose group, The levels of Ros in mitochondria and the content of MDA in cells decreased significantly, while the activities of SOD in cells increased significantly, and the activities of AST and alt in L-02 cells were significantly decreased by high dose quercetin. The level of Ros and the content of MDA in cell mitochondria were significantly increased (P 0.01), and the content of GSH and the activity of main antioxidant enzymes in L-02 cells were significantly increased (P 0.01). The protective effect of high dose quercetin group was more obvious than that of quercetin low dose group. It can be seen that oxidative stress mediated by Ros plays an important role in the cytotoxicity of L-02 cells induced by INH, and quercetin has a protective effect on the oxidative damage of L-02 cells induced by INH.
