芹菜素對雄性大鼠生殖系統(tǒng)功能的影響及相關機制探討

【摘要】:目的研究芹菜素對雄性大鼠生殖系統(tǒng)功能的影響并探討相關機制,為芹菜素的進一步開發(fā)利用提供科學依據(jù)。方法48只SD成年雄性大鼠隨機分為對照組(生理鹽水10ml/kg-bw)和芹菜素低劑量組(234 mg/kg-bw)、中劑量組(468 mg/kg-bw)、高劑量組(936mg/kg·bw)。1次/天,6天/周,連續(xù)5周給予動物生理鹽水或芹菜素灌胃。(1)處死動物,分離睪丸、附睪、精囊腺、前列腺、尿道球腺、肛提肌/球海綿體肌稱重,計算臟器系數(shù)。(2)游離附睪中的精子,采用WLJY-9000系統(tǒng)分析精子密度、運動參數(shù)。制備精子伊紅染色涂片,鏡下觀察精子形態(tài),計算精子畸形率。(3)制備睪丸HE染色病理切片和電鏡切片,觀察睪丸組織結構和細胞超微結構的變化。(4)制備睪丸單細胞懸液,流式細胞儀檢測睪丸細胞周期。(5)制備睪丸組織勻漿,檢測睪丸氧化還原指標T-AOC、SOD、CAT、GSH-Px、GSH、MDA及睪丸組織ACP、AKP、LDH、SDH、NOS、NADPH和Na+K+-ATP酶、Ca2+Mg2+-ATP酶。(6)在灌胃的第7、14、21、28、35天活體心內取血,測定血清T、E2、ICSH、FSH水平。結果(1)各劑量組的睪丸、附睪、陰莖、精囊腺、前列腺、尿道球腺、肛提肌/球海綿體肌的濕重和臟器系數(shù)與對照組比較,差異無統(tǒng)計學意義(P0.05)。(2)高劑量組a級精子密度低于對照組,d級精子密度高于對照組(P0.01);高劑量組精子運動參數(shù)VCL. VSL低于對照組,差異有統(tǒng)計學意義(P0.01)。高劑量組精子畸形率高于對照組,差異有統(tǒng)計學意義(P0.05),畸形類型主要為尾折疊。(3)低、中、高劑量組睪丸組織結構呈不同程度的損傷:生精小管結構不規(guī)則,間質變。簧掀ぷ儽,細胞層數(shù)減少,細胞排列混亂,管腔內成熟精子數(shù)目減少;長形精子細胞出現(xiàn)于生精上皮各層細胞中間。電鏡下觀察到,高劑量組精子尾部線粒體腫脹、線粒體嵴變形。(4)各劑量組睪丸G0/G1期、S期和G2/M期細胞與對照組比較,差異無統(tǒng)計學意義(P0.05)。(5)低、中劑量組T-AOC活力高于對照組,高劑量組T-AOC活力低于對照組(P0.05),低劑量組MDA含量低于對照組(P0.05),低劑量組SOD活力、中劑量組GSH-Px活力低于對照組(P0.05),低、中、高劑量組GSH含量均低于對照組(P0.05)。低、中、高劑量組ACP、AKP和LDH活力均低于對照組(P0.01),低、中、高劑量組Ca2+Mg2+-A-TP酶活力和高劑量組Na+K+-ATP酶活力高于對照組(P0.05)。(6)中、高劑量組T濃度在14天時高于對照組,差異有統(tǒng)計學意義(P0.05);高劑量組E2濃度在21天時低于對照組,差異有統(tǒng)計學意義(P0.05);中劑量組T/E2在28天時高于對照組,高劑量組T/E2在14天和21天時高于對照組(P0.05)。低、高劑量組在28天時,血清ICSH濃度高于對照組(P0.05);各劑量組血清FSH濃度在各時間點與對照組比較,差異無統(tǒng)計學意義(P0.05)。結論(1)936 mg/kg·bw AP降低大鼠精子質量,降低精子運動速度,升高精子畸形率,損傷睪丸組織結構和精子尾部線粒體超微結構。(2)AP對睪丸組織的氧化損傷和能量代謝的干擾可能是精子質量下降的原因。(3)AP可引起血清性激素水平變化,其機制及其對精子質量的影響還需進一步研究。
[Abstract]:Objective To study the effects of apigenin on the reproductive system function of male rats and to explore the related mechanism to provide scientific basis for the further exploitation and utilization of apigenin . Methods 48 SD adult male rats were randomly divided into control group ( normal saline 10ml / kg - bw ) and apigenin low - dose group ( 234 mg / kg - bw ) . Results ( 1 ) The levels of serum T , E2 , ICSH and FSH were significantly lower than those in control group ( P0.05 ) .
High dose group sperm motility parameter VCL . There was significant difference in sperm deformity rate in high - dose group than in control group ( P0.05 ) . The difference was statistically significant ( P0.05 ) .
The number of mature sperms was decreased and the number of mature sperms in the lumen was decreased .
The activity of GSH - Px in the low - dose and medium - dose groups was lower than that in the control group ( P0.05 ) . The activity of the low - dose group and the high - dose group were lower than those in the control group ( P0.05 ) .
The concentration of E2 in high dose group was lower than that of control group at 21 days ( P0.05 ) .
The concentration of T / E2 in the middle dose group was higher than that of the control group at 28 days , and the T / E2 in the high dose group was higher than that of the control group ( P0.05 ) at the 14th day and the 21st day . The serum ICSH concentration was higher in the high dose group than the control group ( P0.05 ) .
Conclusion ( 1 ) 936 mg / kg 路 bw AP can decrease sperm quality in rats , decrease sperm motility , increase sperm deformity rate , damage testis structure and ultrastructure of sperm tail mitochondria .
