神經(jīng)病靶酯酶在三鄰甲苯基磷酸酯抑制小鼠精子發(fā)生的作用

【摘要】:目的:三鄰甲苯磷酸酯(TOCP)常作為增塑劑、軟化劑及阻燃劑等,在工業(yè)中使用非常廣泛,本課題主要觀察TOCP對小鼠睪丸曲細精管及精子發(fā)生的影響,,探究神經(jīng)病靶酯酶(NTE)在TOCP抑制小鼠精子發(fā)生中的作用。 方法:1、用HE染色法觀察TOCP對小鼠睪丸曲細精管及精子發(fā)生的影響;2、用免疫組織化學方法檢測NTE在小鼠睪丸中表達;3、用戊酸苯酯水解法檢測TOCP對小鼠睪丸NTE活性的影響4、用RT-PCR法檢測TOCP對小鼠睪丸組織中NTE mRNA表達的影響。 結(jié)果:1、分別用不同濃度(0,100,200,400mg/kg)的TOCP給成年雄性小鼠連續(xù)染毒14d后,結(jié)果顯示,與對照組相比,200和400mg/kg TOCP處理組小鼠的睪丸系數(shù)明顯降低(P<0.05),而100mg/kg濃度的TOCP則并沒有明顯抑制小鼠的睪丸系數(shù)(P0.05)。不同濃度的TOCP對附睪系數(shù)則無明顯差異(P0.05)。2、與對照組相比,200和400mg/kg TOCP處理組小鼠睪丸的曲細精管結(jié)構(gòu)顯著紊亂,其生精細胞脫落,而100mg/kg TOCP處理組小鼠的睪丸曲細精管則無明顯破壞。與對照組相比,100,200及400mg/kg的TOCP均可抑制精子的數(shù)量(P<0.05)。3、免疫組織化學結(jié)果顯示,除曲細精管的表層細胞之外,NTE在曲細精管的不同生精細胞(包括精原干細胞、初級精母細胞、次級精母細胞和精子)均高表達。4、TOCP呈濃度依賴式地抑制小鼠睪丸中的NTE活性(P<0.05),但對小鼠睪丸中的NTE mRNA表達無明顯的影響(P0.05)。 結(jié)論:1、TOCP降低睪丸系數(shù)而對附睪系數(shù)無明顯影響;2、TOCP明顯損傷睪丸的曲細精管并抑制精子發(fā)生;3、NTE在小鼠睪丸組織中表達;4、TOCP抑制睪丸中NTE活性而對其mRNA表達無明顯影響。
[Abstract]:Objective: Trio-toluene phosphate (TOCP) is widely used in industry as plasticizer, softener and flame retardant. The effect of TOCP on testicular seminiferous tubules and spermatogenesis in mice was observed. To explore the role of neuropathic target esterase (NTE2) in the inhibition of spermatogenesis in mice by TOCP. Methods: 1. The effect of TOCP on testicular seminiferous tubules and spermatogenesis in mice was observed by HE staining, and NTE was detected by immunohistochemistry in testis of mice. The effect of TOCP on the activity of NTE in testis of mice was detected by phenyl valerate hydrolysis. (4) the effects of TOCP on the expression of NTE mRNA in testis of mice were detected by RT-PCR. The results showed that compared with the control group, the testicular coefficient of mice treated with TOC 200 and 400mg/kg was significantly lower than that of the control group (P < 0.05), while the TOC with 100mg/kg concentration did not significantly inhibit the testicular coefficient of mice (P 0.05). Compared with the control group, the testicular seminiferous tubules of the mice treated with different concentrations of TOCP were significantly disordered and spermatogenic cells were shedded, but the testicular seminiferous tubules of the mice treated with 100mg/kg TOCP had no obvious damage to the seminiferous tubules. Compared with the control group, TOC of TOC 100 200 and 400mg/kg could inhibit the number of spermatozoa (P < 0 05. 3). The immunohistochemical results showed that NTE was present in different spermatogenic cells (including spermatogonial stem cells and primary spermatocytes) in the seminiferous tubules except the surface cells of the seminiferous tubules. Both secondary spermatocytes and spermatozoa) were highly expressed. (4) TOCP inhibited NTE activity in mouse testis in a concentration-dependent manner (P < 0.05), but had no significant effect on NTE mRNA expression in testis of mice (P < 0.05). Conclusion: TOC can decrease testicular coefficient but not epididymis coefficient. TOC significantly affected the damage of testicular tubules and inhibited spermatogenesis in testicular tissues of mice. TOC inhibited the activity of NTE in testis, but had no effect on its mRNA expression.
