磷酸三苯酯對青春期雄性SD大鼠睪丸增殖細(xì)胞核抗原和芳香化酶表達(dá)的影響

【摘要】:目的: 觀察磷酸三苯酯對青春期大鼠睪丸組織的影響,及其對相關(guān)基因和蛋白表達(dá)的影響,初步研究磷酸三苯酯對大鼠生殖系統(tǒng)的作用。 方法: 將40只健康的23日齡SPF級雄性SD大鼠隨機(jī)分為4組,每組10只,即溶劑對照組,低劑量組(250mg/kg)、中劑量組(500mg/kg)、高劑量組(750mg/kg);配成相應(yīng)的玉米油溶液,按0.5%體重灌胃。連續(xù)灌胃染毒30天后,取大鼠雙側(cè)睪丸組織,運(yùn)用HE染色觀察各實驗組大鼠睪丸組織的病理改變;免疫組織化學(xué)染色觀察各組大鼠睪丸PCNA和AROM蛋白的表達(dá);應(yīng)用RT-PCR檢測各組大鼠睪丸PCNA、 AROM mRNA的表達(dá):應(yīng)用Western blot檢測各組大鼠睪丸PCNA、 AROM蛋白的表達(dá)。 結(jié)果: 與對照組相比,染毒組大鼠的體重明顯下降(P0.05),而四個組相應(yīng)的睪丸臟器系數(shù)無統(tǒng)計學(xué)意義(P0.05)。HE染色顯示:對照組大鼠的生精小管結(jié)構(gòu)正常,各級生精細(xì)胞排列有序。各染毒組可見生精上皮發(fā)生變性,各級生精細(xì)胞排列紊亂,甚至出現(xiàn)了精子扭曲纏結(jié),腫脹,脫落的現(xiàn)象(P0.05)。免疫組化檢測結(jié)果顯示,PCNA、AROM在染毒劑量組睪丸組織的陽性表達(dá)強(qiáng)度降低,與對照組比較其差別有顯著性(P0.05)。RT-PCR結(jié)果表明,各劑量組大鼠睪丸組織的PCNA、 AROM mRNA表達(dá)降低,與對照組相比具有統(tǒng)計學(xué)意義(P0.05)。Western blot結(jié)果顯示,各劑量組大鼠睪丸的PCNA、AROM蛋白表達(dá)明顯降低,與對照組相比具有統(tǒng)計學(xué)意義(P0.05)。 結(jié)論: TPP能抑制大鼠睪丸PCNA、AROM mRNA和蛋白的表達(dá),提示雄性大鼠青春期暴露于TPP,可以通過損傷睪丸組織結(jié)構(gòu),降低PCNA、 AROM表達(dá)水平等機(jī)制,從而對大鼠的睪丸發(fā)育產(chǎn)生損害。
[Abstract]:Objective: To observe the effect of triphenyl phosphate on testis of pubertal rats, and to study the effect of triphenyl phosphate on the reproductive system of rats. Methods: Forty healthy 23-day-old male SD rats of SPF grade were randomly divided into 4 groups, 10 rats in each group: solvent control group, low dose group (250 mg / kg), middle dose group (500 mg / kg), high dose group (750 mg / kg), and corresponding corn oil solution (0.5% body weight). After 30 days of intragastric administration, the testis of the rats were collected and the pathological changes of testis in each experimental group were observed by HE staining, and the expressions of PCNA and AROM protein in testis of each group were observed by immunohistochemical staining. RT-PCR was used to detect the expression of PCNA, AROM mRNA in testis of rats and Western blot was used to detect the expression of PCNA and AROM in testis. Results: Compared with the control group, the body weight of the rats in the exposed group decreased significantly (P 0.05A), while the viscera coefficients of the testis in the four groups were not statistically significant (P 0.05). He staining showed that the structure of the seminiferous tubules in the control group was normal, and the spermatogenic cells of the four groups were arranged in an orderly manner. The spermatogenic epithelium was denatured, the spermatogenic cells of all levels were disordered, and the spermatozoa were twisted and tangled, swelled and shedded (P0.05). The results of immunohistochemistry showed that the positive expression intensity of PCNA, AROM mRNA in testicular tissue of the exposed group was lower than that of the control group. The results of RT-PCR showed that the expression of PCNA, AROM mRNA in the testis of each dose group was lower than that of the control group. Compared with the control group, the results of blot showed that the expression of PCNAA-AROM in testis of each dose group was significantly lower than that of the control group (P 0.05), and that of the control group was significantly lower than that of the control group (P 0.05). Conclusion: TPP can inhibit the expression of PCNAA-AROM mRNA and protein in testis of rats, suggesting that the mechanism of male rats exposed to TPP during puberty may damage the testis development by damaging the structure of testis and decreasing the expression level of AROM and so on.
