【摘要】：目的探討鋁對(duì)大鼠腎上腺嗜鉻細(xì)胞瘤(PC12)細(xì)胞β-淀粉樣蛋白(Aβ)42表達(dá)的影響及其作用機(jī)制。方法將處于對(duì)數(shù)生長(zhǎng)期的PC12細(xì)胞分別暴露于含麥芽酚鋁終濃度為0(對(duì)照)、50、100、200和400μmol/L的完全培養(yǎng)基染毒12、24、48 h。采用CCK-8法測(cè)定細(xì)胞活性,采用酶聯(lián)免疫吸附(ELISA)測(cè)定法測(cè)定PS1、Aβ42蛋白的含量。結(jié)果與對(duì)照組比較,200、400μmol/L麥芽酚鋁染毒12 h時(shí)及各濃度麥芽酚鋁染毒24 h、48 h時(shí)PC12細(xì)胞的存活率均較低,差異有統(tǒng)計(jì)學(xué)意義(P0.05);且隨著麥芽酚鋁染毒濃度的升高,不同染毒時(shí)間PC12細(xì)胞的存活率均呈逐漸降低的趨勢(shì)。與對(duì)照組比較,400μmol/L麥芽酚鋁染毒12 h時(shí)及200、400μmol/L麥芽酚鋁染毒24 h、48 h時(shí)PC12細(xì)胞PS1蛋白的含量均增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。與對(duì)照組比較,400μmol/L麥芽酚鋁染毒12 h時(shí)及200、400μmol/L麥芽酚鋁染毒24 h時(shí)以及100、200、400μmol/L麥芽酚鋁染毒48 h時(shí)PC12細(xì)胞Aβ42蛋白的含量均增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論γ-分泌酶表達(dá)增加可能是鋁致PC12細(xì)胞Aβ42蛋白含量增多的重要原因之一。
[Abstract]:Objective to investigate the effect of aluminum on the expression of 尾 -amyloid protein A 尾 -chl42 in rat adrenal pheochromocytoma (PC12) cells and its mechanism. Methods PC12 cells at logarithmic growth stage were exposed to a complete medium containing aluminum-maltol at 0 (control group 50100200 and 400 渭 mol/L) for 48 h. The cell activity was determined by CCK-8 and the protein content of PS1 A 尾 42 was determined by Elisa. Results compared with the control group, the survival rate of PC12 cells was lower at 12 h and 24 h and 48 h, respectively, and the difference was statistically significant (P 0.05), and with the increase of maltol aluminum concentration, the survival rate of PC12 cells was significantly lower than that of control group (P < 0.05), and with the increase of maltol aluminum concentration, the survival rate of PC12 cells was significantly lower than that of control group (P < 0.05). The survival rate of PC12 cells decreased gradually at different exposure time. Compared with the control group, the content of PS1 protein in PC12 cells increased at 12 h and 200400 渭 mol/L, 24 h and 48 h, respectively, and the difference was statistically significant (P 0.05). Compared with the control group, the A 尾 42 protein content of PC12 cells increased at 12 h, 200400 渭 mol/L, 24 h and 48 h, respectively, and the difference was statistically significant (P 0.05). Conclusion the increased expression of 緯 -secretase may be one of the important reasons for the increase of A 尾 42 protein content in PC12 cells induced by aluminum.
