【摘要】:目的:探討新疆雙峰駝乳清蛋白組分對人宮頸癌HeLa細(xì)胞增殖的抑制作用.方法:采用四甲基偶氮唑鹽(MTT)法測定新疆雙峰駝乳蛋白組分對HeLa細(xì)胞增殖的抑制作用;熒光染色檢測HeLa細(xì)胞凋亡的形態(tài)變化;倒置顯微鏡、掃描電鏡和透射電鏡觀察HeLa細(xì)胞的形態(tài)變化;用流式細(xì)胞術(shù)檢測HeLa細(xì)胞凋亡率、線粒體膜電位的變化.結(jié)果表明:新疆雙峰駝乳清蛋白組分TR35可抑制人宮頸癌HeLa細(xì)胞的增殖并誘導(dǎo)其凋亡,并呈時間和劑量依賴性(P0.01);倒置顯微鏡、掃描電鏡和透射電鏡觀察到典型的凋亡特征,熒光顯微觀察到凋亡小體;Annexin-Ⅴ/PI雙染檢測出現(xiàn)早期凋亡細(xì)胞;線粒體膜電位去極化,并將細(xì)胞周期阻滯在S期.結(jié)論:新疆雙峰駝乳清蛋白組分TR35可抑制HeLa細(xì)胞的增殖和誘導(dǎo)細(xì)胞凋亡,TR35組分誘導(dǎo)細(xì)胞凋亡的途徑可能與線粒體通路有關(guān).
[Abstract]:Aim: to investigate the inhibitory effect of whey protein on the proliferation of human cervical cancer HeLa cells in Xinjiang. Methods: the inhibitory effect of lactoprotein on the proliferation of HeLa cells was determined by tetramethylazolium tetrazolium (MTT) method. The morphological changes of apoptosis of HeLa cells were detected by fluorescence staining. The morphological changes of HeLa cells were observed by scanning electron microscope and transmission electron microscope, and the apoptosis rate and mitochondrial membrane potential of HeLa cells were detected by flow cytometry. The results showed that the whey protein component TR35 could inhibit the proliferation and induce apoptosis of human cervical cancer HeLa cells in a time-and dose-dependent manner, and the typical apoptotic characteristics were observed by inverted microscope, scanning electron microscope and transmission electron microscope.
