敵敵畏對腎細(xì)胞系293T增殖的影響

【摘要】:為了研究敵敵畏(dichlorvos,DDVP)對人腎細(xì)胞的毒性效應(yīng)。應(yīng)用MTT比色法檢測敵敵畏對人腎細(xì)胞系293T細(xì)胞增殖的影響,流式細(xì)胞儀檢測293T細(xì)胞的凋亡情況,熒光二氯熒光素二乙酸(DCF-DA)測定細(xì)胞內(nèi)的活性氧(reactive oxidative species,ROS)水平。結(jié)果表明:用濃度為0.5 mmol/L、1 mmol/L、5 mmol/L、10 mmol/L、15 mmol/L和20 mmol/L的敵敵畏分別處理293T細(xì)胞1 h后發(fā)現(xiàn)敵敵畏對293T的生長抑制具有劑量效應(yīng),計算得到1 h的IC_(50)值為10.406 mmol/L;不同濃度敵敵畏(0.5 mmol/L,5 mmol/L,10 mmol/L)處理1 h 293T細(xì)胞發(fā)現(xiàn)細(xì)胞產(chǎn)生明顯的凋亡現(xiàn)象,隨著處理濃度的增加,細(xì)胞的凋亡率升高,檢測細(xì)胞內(nèi)活性氧水平發(fā)現(xiàn)細(xì)胞內(nèi)活性氧水平也隨之升高。結(jié)果提示敵敵畏能夠通過凋亡途徑抑制293T細(xì)胞的增殖,其作用機(jī)制可能是通過增加293T細(xì)胞內(nèi)活性氧水平,促進(jìn)細(xì)胞凋亡,進(jìn)而抑制細(xì)胞增殖。
[Abstract]:To study the toxic effect of dichlorvosine DDVP on human renal cells. The effects of dichlorvos on the proliferation of human renal cell line 293T were detected by MTT colorimetry, apoptosis of 293T cells was detected by flow cytometry, and reactive oxidative speciesros were measured by fluorescence dichlorofluorescein diacetate (DCF-DAA). The results showed that dichlorvos at the concentration of 0.5 mmol / L ~ (-1) 5 mmol / L ~ (10) mmol 路L ~ (-1) and 10 mmol / L ~ (15) mmol/L and 20 mmol/L, respectively, were used to treat 293T cells for 1 h, and the results showed that dichlorvos had a dose effect on the growth inhibition of 293T cells. After 1 h treatment with 0.5 mmol / L dichlorvos (0.5 mmol / L) and 10 mmol / L (10 mmol / L) of dichlorvos, it was found that the apoptosis of 293T cells was obvious with the increase of the concentration of dichlorvos, and the apoptosis rate of the cells increased with the increase of the concentration of dichlorvos. Detection of intracellular reactive oxygen species showed that intracellular reactive oxygen species levels also increased. The results suggested that dichlorvos could inhibit the proliferation of 293T cells through the apoptotic pathway, and its mechanism might be to promote cell apoptosis by increasing the level of reactive oxygen species in 293T cells and thus inhibit cell proliferation.
