水體細(xì)菌微生物多波長透射光譜快速準(zhǔn)確獲取方法研究

【摘要】:為實現(xiàn)水體細(xì)菌微生物快速在線監(jiān)測,搭建了多波長透射光譜快速測量實驗系統(tǒng),利用該系統(tǒng)分別測量了重鉻酸鉀標(biāo)準(zhǔn)溶液紫外波段及中性濾光片可見波段的透射光譜,并與紫外-可見分光光度計測得的透射光譜進行對比分析,驗證了實驗系統(tǒng)測量透射光譜的準(zhǔn)確性;以水體中常見的金黃色葡萄球菌作為研究對象,利用搭建的實驗系統(tǒng)獲取金黃色葡萄球菌溶液在220~900nm波段的前向小角度透射光譜,進一步驗證了實驗系統(tǒng)測量細(xì)菌微生物透射光譜的準(zhǔn)確性和快速性。結(jié)果表明,由實驗系統(tǒng)和紫外-可見分光光度計測得的重鉻酸鉀標(biāo)準(zhǔn)溶液,與中性濾光片紫外波段及可見波段透射光譜的線性擬合相關(guān)系數(shù)分別為0.999 7和0.999 5,光密度誤差分別在5.00%和4.58%以內(nèi),說明兩個系統(tǒng)測量光譜的一致性較好,所搭建的實驗系統(tǒng)測量標(biāo)準(zhǔn)樣品紫外-可見透射光譜準(zhǔn)確度較高;對于金黃色葡萄球菌,實驗系統(tǒng)測得的透射光譜經(jīng)過校正后,與紫外-可見分光光度計測得的透射光譜線性擬合的相關(guān)系數(shù)為0.999 97,兩者相比的光密度誤差在0.74%以內(nèi);系統(tǒng)重復(fù)30次細(xì)菌光譜信號采集獲得平均透射光譜單次測量時間為15s,說明該實驗系統(tǒng)相對于紫外-可見分光光度計能夠快速準(zhǔn)確獲取水體細(xì)菌微生物多波長透射光譜,在保證測量結(jié)果準(zhǔn)確的同時縮短了光譜測量時間,為水體細(xì)菌微生物快速檢測提供技術(shù)支持。
[Abstract]:In order to realize the rapid on-line monitoring of bacteria and microbes in water body, a multi-wavelength transmission spectrum fast measurement system was set up. The transmission spectra of ultraviolet band and neutral filter band of potassium dichromate standard solution were measured by the system. Compared with the transmission spectrum measured by the UV-Vis spectrophotometer, the accuracy of the experimental system was verified, and the common staphylococcus aureus in water was used as the research object. The low angle forward transmission spectra of Staphylococcus aureus solution in 220~900nm band were obtained by using the established experimental system, which further verified the accuracy and rapidity of the experimental system in measuring the transmission spectrum of bacteria microbes. The results show that the standard solution of potassium dichromate obtained from the experimental system and UV-Vis spectrophotometer, The linear fitting correlation coefficient with the transmission spectrum in the ultraviolet and visible bands of the neutral filter is 0.999 7 and 0.999 5, respectively, and the optical density error is less than 5.00% and 4.58%, respectively, which indicates that the measurement spectra of the two systems are in good agreement. The experimental system has high accuracy in measuring the UV-Vis transmission spectra of standard samples, and for Staphylococcus aureus, the transmission spectra measured by the experimental system have been corrected. The correlation coefficient of linear fitting of transmission spectrum measured by UV-Vis spectrophotometer is 0.999, and the optical density error is less than 0.74%. The average time of single measurement of transmission spectrum is 15 s, which indicates that the system can quickly and accurately obtain the multiwavelength transmission spectrum of bacteria and microorganism in water compared with the UV-Vis spectrophotometer. At the same time, it can shorten the time of spectrum measurement and provide technical support for rapid detection of bacteria and microorganisms in water body.
