四氯化碳聯(lián)合二乙基亞硝胺對(duì)小鼠精巢的毒性作用

【摘要】:為了研究四氯化碳(CCL4)聯(lián)合二乙基亞硝胺(DEN)對(duì)小鼠(Mus musculus domesticus)精巢的毒性作用。將昆明小鼠隨機(jī)分為對(duì)照組與實(shí)驗(yàn)組,各12只個(gè)體。實(shí)驗(yàn)組每周2次腹腔注射含有15%CCl4的花生油溶液,同時(shí)自由飲用0.07‰的DEN溶液,連續(xù)誘導(dǎo)8周。對(duì)照組每周2次注射花生油,飲用蒸餾水。分別于第3、6、8周處死實(shí)驗(yàn)組及對(duì)照組小鼠各4只,對(duì)其精巢組織進(jìn)行常規(guī)固定、包埋、切片、H.E染色,觀察精巢的形態(tài)及精子數(shù)量的變化。與實(shí)驗(yàn)組比較,對(duì)照組小鼠體重明顯增加,且實(shí)驗(yàn)組與對(duì)照組差異顯著;與對(duì)照組相比,實(shí)驗(yàn)組睪丸系數(shù)以及精子存活率均顯著減少。對(duì)照組小鼠精巢都具有正常的生精小管結(jié)構(gòu),管腔中存在大量精子;第3周,實(shí)驗(yàn)組小鼠的精巢與對(duì)照組類似,生精小管較為完整,生精細(xì)胞稍有散亂;第6周,實(shí)驗(yàn)組生精小管變形,生精細(xì)胞排列疏松,精子數(shù)目減少且畸形;第8周,實(shí)驗(yàn)組生精小管殘缺不全,生精細(xì)胞和精子散亂排列,精子數(shù)量大大減少且變形更為嚴(yán)重。實(shí)驗(yàn)說(shuō)明,CCl4聯(lián)合DEN能夠造成小鼠精巢組織的損害以及精子數(shù)目的減少和畸變。
