【摘要】：[目的]研究飼料鈣對(duì)母鼠飲水型氟染毒后子代大鼠腎細(xì)胞線粒體損傷的影響。[方法]選用健康初斷乳SD雌性大鼠100只,隨機(jī)分為對(duì)照組、染氟組(100 mg/L Na F)、低鈣組(0.063%CaCO_3)、低鈣染氟組(100 mg/L Na F+0.063%CaCO_3)和高鈣染氟組(100 mg/L Na F+7%CaCO_3);飼養(yǎng)3個(gè)月后,雌雄鼠合籠交配。取日齡14 d及28 d仔鼠雌雄各10只,以其腎臟細(xì)胞線粒體標(biāo)志酶琥珀酸脫氫酶(SDHase)活性及脂質(zhì)過(guò)氧化指標(biāo)丙二醛(MDA)水平,腎臟細(xì)胞凋亡狀況,線粒體分裂/融合蛋白Fis1、Drp1和Mfn2表達(dá)水平為觀察指標(biāo)。[結(jié)果]與染氟組相比,高鈣染氟組SDHase活性升高(P0.05),低鈣染氟組SDHase活性降低(P0.05)。與對(duì)照組相比,雌鼠各組腎臟線粒體MDA含量均升高(P0.05)。與對(duì)照組相比,染氟各組仔鼠凋亡細(xì)胞增多;與染氟組相比,低鈣染氟組凋亡細(xì)胞增多,而高鈣染氟組凋亡細(xì)胞減少。與對(duì)照組相比,低鈣染氟組14 d雄鼠的分裂蛋白Fis1表達(dá)升高(P0.05);低鈣染氟組和高鈣染氟組28 d雄鼠的分裂蛋白Drp1升高(P0.05)。[結(jié)論]氟中毒能夠造成大鼠腎臟細(xì)胞線粒體內(nèi)分裂/融合蛋白Fis1、Drp1和Mfn2表達(dá)異常,引起腎臟細(xì)胞線粒體損傷。高鈣飼料攝入能降低線粒體內(nèi)脂質(zhì)過(guò)氧化反應(yīng),減輕高氟對(duì)子代腎臟細(xì)胞的毒性作用,而低鈣飼料攝入會(huì)加劇高氟的毒性作用。
[Abstract]:[objective] to study the effect of dietary calcium on mitochondria damage of rat renal cells after drinking fluoride exposure. [methods] 100 healthy female SD rats were randomly divided into three groups: control group (n = 100), 0.063CO_3 group (n = 100) and high Ca ~ (2 +) -exposed group (n = 100 mg/L Na F 7CO_3). Females and males mate in cages. The activity of SDHase and the level of malondialdehyde (MDAs), a marker of succinate dehydrogenase (SDHase) and lipid peroxidation, were measured in 10 male and 10 male rats of 14 d and 28 d, respectively, and the apoptosis of renal cells was observed. The expression levels of Drp1 and Mfn2 in mitochondrial division / fusion protein Fis1 were observed. [results] compared with the fluoride group, the SDHase activity of the high calcium fluoride group was higher than that of the low calcium fluoride group, while the SDHase activity of the low calcium fluoride group was lower than that of the low calcium fluoride group. Compared with the control group, the content of MDA in kidney mitochondria of female rats was higher than that of control group. Compared with the control group, the number of apoptotic cells in fluoride exposed group was higher than that in fluoride group, but the number of apoptotic cells in low calcium fluoride group was higher than that in high calcium fluoride group. Compared with the control group, the expression of mitogen Fis1 in the low calcium fluoride group was higher than that in the control group on the 14th day, and that in the low calcium fluoride group and the high calcium fluoride group at 28 days was higher than that in the control group. [conclusion] fluoride poisoning can cause abnormal expression of Dr p1 and Mfn2 in mitochondria of rat kidney cells, and cause mitochondrial damage of renal cells. High calcium diet can reduce lipid peroxidation in mitochondria and reduce the toxicity of high fluoride to kidney cells of offspring, but low calcium diet can aggravate the toxic effect of high fluoride.
