靈芝孢子粉對(duì)亞慢性染鎘大鼠肝臟損傷的保護(hù)作用

【摘要】:目的:研究靈芝孢子粉對(duì)亞慢性染鎘大鼠肝臟損傷的保護(hù)作用,并探討其可能的保護(hù)途徑。方法:健康雄性SD大鼠84只,體重(90±10)g,隨機(jī)分為:正常對(duì)照組(C)、單獨(dú)染鎘組(Cadmium,Cd)和2個(gè)給藥劑量組(靈芝孢子粉0.5g/kg,1.0g/kg+Cd Cl2,即GCd L和GCdH組),每組各21只。大鼠適應(yīng)性喂養(yǎng)7d后,每天對(duì)GCdL組和GCdH組大鼠灌相應(yīng)劑量的靈芝孢子粉,C組和Cd組灌等體積蒸餾水。Cd組、GCdL組和GCd H組每隔一天腹腔注射一次氯化鎘(1.0mg/kg),每周稱重1次,并觀察大鼠的一般情況,并在第30天、60天、90天分批處死,每組各處死7只;頸動(dòng)脈取血,快速取肝臟,生理鹽水沖洗,濾紙吸干多余的水分,稱重,記錄,-80℃保存;全自動(dòng)血生化儀檢測(cè)大鼠丙氨酸氨基轉(zhuǎn)移酶(Alanine aminotransferase,ALT)和天門冬氨酸氨基轉(zhuǎn)移酶(Aspartate transaminase,AST)的濃度;光學(xué)顯微鏡觀察大鼠肝臟病理組織學(xué)變化;比色法檢測(cè)肝臟丙二醛(Malondialdehyde,MDA)含量;酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測(cè)大鼠肝臟IL-6含量;Real time RT-PCR檢測(cè)大鼠肝臟p65 m RNA、Mt-1 m RNA、Mt-2 m RNA的表達(dá)量。結(jié)果:與正常對(duì)照組比較,60d、90d的Cd組大鼠體重明顯降低(P0.05),給予靈芝孢子粉干預(yù)后,大鼠體重均有不同程度增加,在90天時(shí),GCdH組大鼠體重明顯高于Cd組(P0.05);與空白組相比,大鼠染鎘后肝臟臟器系數(shù)明顯增大(P0.05),給藥劑量組大鼠肝臟臟器系數(shù)與Cd組比較均有不同程度降低(P0.05);與空白組相比,單獨(dú)染鎘組ALT、AST明顯升高(P0.05),而靈芝孢子粉各劑量組ALT、AST均有不同程度降低,且與單獨(dú)染鎘組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);HE染色結(jié)果顯示,單獨(dú)染鎘組肝組織細(xì)胞間質(zhì)充血嚴(yán)重,細(xì)胞排列雜亂,細(xì)胞界限不清晰,而給藥組以上情況均有不同程度改善,并且高劑量組改善情況優(yōu)于中劑量組;比色法檢測(cè)MDA結(jié)果顯示,經(jīng)鎘處理大鼠肝臟MDA含量較空白組明顯增高(P0.05),給予靈芝孢子粉干預(yù)后,在60d、90d均有降低(P0.05);在30、60、90天,與空白組相比,單獨(dú)染鎘組大鼠肝臟細(xì)胞因子IL-6(白介素6)含量明顯升高(P0.05);隨著時(shí)間的延長,Cd組、GCdL組、GCdH組IL-6含量也逐漸增加(P0.05);與單獨(dú)染鎘組相比,給藥組大鼠肝臟Mt-1 m RNA、Mt-2 m RNA的表達(dá)量明顯升高(P0.05),而p65 m RNA表達(dá)明顯降低(P0.05)。結(jié)論:靈芝孢子粉對(duì)亞慢性染鎘大鼠肝臟損傷有保護(hù)作用,其機(jī)制可能與抗氧化應(yīng)激,降低炎性細(xì)胞因子IL-6表達(dá)和誘導(dǎo)MT產(chǎn)生有關(guān)。
[Abstract]:Aim: to study the protective effect of ganoderma lucidum spores powder on liver injury in rats with subchronic cadmium exposure and explore its possible protective pathway. Methods: 84 healthy male Sprague-Dawley rats, weighing 90 鹵10 g / g, were randomly divided into two groups: normal control group, single cadmium group and two dosage groups (Ganoderma lucidum spores 0.5 g / kg 路kg ~ (-1) 路kg ~ (-1) CD _ (Cl _ 2), I. e., GCd L group and GCdH group, 21 rats in each group.
