RP-HPLC法測(cè)定市售綠茶飲料中茶多酚和生物堿的含量

【摘要】:目的建立HPLC同時(shí)測(cè)定市售綠茶飲料中茶多酚和生物堿含量的方法并考察放置時(shí)間對(duì)其產(chǎn)生的影響。方法樣品經(jīng)SPE小柱萃取后進(jìn)樣分析。色譜柱為Phenomemex Synergi 4u FusionRP ODS(250 mm×4.6 mm,5μm),流動(dòng)相為乙腈-水,梯度洗脫,流速為1.0 mL·min~(-1),檢測(cè)波長(zhǎng)為210 nm,柱溫為25℃。結(jié)果在上述色譜條件下,6個(gè)待測(cè)成分在測(cè)定范圍內(nèi)線性關(guān)系良好(r"g0.999 1),平均加樣回收率為97.3%~99.1%,RSD"f2.9%(n=9);隨放置時(shí)間的延長(zhǎng),飲料中茶多酚的含量降低,生物堿含量無(wú)明顯變化。結(jié)論隨放置時(shí)間的延長(zhǎng),飲料中茶多酚的損失率增加,含量減少,其抗氧化活性有所降低。
[Abstract]:Objective to establish a HPLC method for simultaneous determination of tea polyphenols and alkaloids in green tea drinks and to investigate the effect of storage time on the content of tea polyphenols and alkaloids. Methods the samples were extracted by SPE column and analyzed by injection. The chromatographic column was Phenomemex Synergi 4u FusionRP ODS(250 mm 脳 4.6mm ~ 5 渭 m ~ (-1), the mobile phase was acetonitrile-water, gradient elution, the flow rate was 1.0 mL / min ~ (-1), the detection wavelength was 210nm and the column temperature was 25 鈩,
