雙酚AF脅迫對(duì)斑馬魚早期發(fā)育相關(guān)基因的影響

【摘要】:[目的]探討雙酚AF脅迫對(duì)斑馬魚早期發(fā)育的影響。[方法]采用半靜態(tài)染毒的方式暴露受精卵7d(0、5、50和500μg/L),通過(guò)顯微觀察與實(shí)時(shí)熒光定量PCR方法,檢測(cè)孵化率及相關(guān)基因表達(dá)量變化。[結(jié)果]50、500μg/L BPAF暴露影響斑馬魚胚胎72 h孵化率,分別下降44.97%、49.44%;下丘腦trh基因在各暴露組無(wú)明顯變化,而下丘腦crh基因在胚胎(幼魚)期各暴露組分別上調(diào)了44.14%(40.45%)、38.55%(95.17%)、70.65%(83.25%);胚胎期垂體tsh-β基因表達(dá)水平在50、500μg/L濃度組顯著上調(diào),而幼魚期tsh-β基因表達(dá)水平僅在50μg/L暴露組顯著上調(diào)。[結(jié)論]雙酚AF脅迫能夠降低斑馬魚胚胎72 h孵化率,crh基因?qū)PAF脅迫較敏感,最低有影響濃度LOEC為5μg/L。
[Abstract]:[objective] to study the effect of bisphenol A F stress on the early development of zebrafish. [methods] the fertilized eggs were exposed to 50 渭 g / L and 500 渭 g / L of fertilized eggs for 7 days by semi-static exposure. The hatching rate and the expression of related genes were detected by microscopic observation and real-time fluorescence quantitative PCR. [results] 50500 渭 g / L BPAF exposure affected the hatching rate of zebrafish embryos at 72 h, and decreased 44.97% and 49.44%, respectively, while the hypothalamic trh gene did not change significantly in all exposed groups. However, the hypothalamic crh gene was up-regulated by 44.14% and 40.45% in each exposure group, respectively, and the expression level of Tsh- 尾 gene in embryonic pituitary was significantly increased in 50500 渭 g / L group and only in 50 渭 g / L exposure group, and 70.65% in embryo stage, and only in 50 渭 g / L exposure group in immature fish stage, and the expression level of Tsh- 尾 gene was significantly increased in 50 渭 g / L exposure group in embryo stage. The expression of Tsh- 尾 gene in hypothalamus was significantly up-regulated in 50500 渭 g / L group and only in 50 渭 g / L group. [conclusion] Bisphenol A F stress can reduce the hatching rate of zebrafish embryos at 72 h and CRH gene is sensitive to BPAF stress, and the lowest LOEC concentration is 5 渭 g / L.
