納米硫化鎘對A549細(xì)胞的損傷研究

【摘要】:研究納米硫化鎘(Nano-Cd S)材料對肺癌細(xì)胞系A(chǔ)549的毒性及氧化損傷作用。培養(yǎng)A549細(xì)胞,經(jīng)傳代后接種于6孔板中,每孔2 m L完全培養(yǎng)基,接種次日進(jìn)行染毒。用直徑20~30 nm、長度80~100 nm的Nano-Cd S進(jìn)行染毒,染毒濃度分別為0、5、10、20、40和80 mg·L~(-1)。染毒24 h后用MTT檢測細(xì)胞存活率,以存活率在80%左右的濃度為后續(xù)實(shí)驗(yàn)染毒濃度。應(yīng)用流式細(xì)胞技術(shù),用熒光探針法檢測A549細(xì)胞的活性氧(reactive oxygen species,ROS)含量,PI-Annexin-V法檢測細(xì)胞凋亡情況;用試劑盒檢測細(xì)胞中超氧化物岐化酶(superoxide dismutase,SOD)和過氧化氫酶(catalase,CAT)活性以及丙二醛(malondialdehyde,MDA)含量,判斷細(xì)胞氧化損傷情況。不同濃度Nano-Cd S處理細(xì)胞24 h之后,細(xì)胞存活率隨劑量的增加而下降,濃度為10、20、40和80μg·L~(-1)時,存活率分別為(88.71%±0.80%)、(81.93%±3.06%)、(75.23%±1.13%)和(70.66%±5.63%),且各組間差異均具有統(tǒng)計(jì)學(xué)意義(P0.05)。以濃度為10和20 mg·L~(-1)的Nano-Cd S染毒24 h后,胞內(nèi)ROS含量和細(xì)胞凋亡率隨染毒劑量的增加而增加(P0.05);濃度為10 mg·L~(-1)時,細(xì)胞凋亡率為(6.26%±0.44%)。與對照相比,各染毒組SOD和CAT活性和MDA含量升高,20 mg·L~(-1)染毒組SOD和CAT活性和MDA含量高于10 mg·L~(-1)染毒組(P0.05)。研究表明,納米硫化鎘能引起A549細(xì)胞的氧化損傷和細(xì)胞凋亡,具有明顯的細(xì)胞毒性。
[Abstract]:The toxicity and oxidative damage effect of nano cadmium sulfide (Nano-Cd S) on lung cancer cell line A549 was studied. A549 cells were cultured and inoculated into 6 orifice plates after subpassages, 2 m L per pore, and infected on the next day. Nano-Cd S, diameter 20~30 nm, and 80~100 nm length were poisoned respectively. After 24 h, the cell survival rate was detected by MTT and the concentration of the survival rate was about 80%. The flow cytometry was used to detect the content of reactive oxygen species (ROS) in A549 cells by fluorescence probe, and the cell apoptosis was detected by PI-Annexin-V method, and the superoxide dismutase (SOD) in the cell was detected by the kit. Superoxide dismutase, SOD) and the activity of catalase (catalase, CAT) and the content of malondialdehyde (malondialdehyde, MDA) were used to determine the oxidative damage of cells. After 24 h of Nano-Cd S treated cells with different concentrations, the survival rate of cells decreased with the increase of dosage, and the survival rate was (88.71% + 0.80%), respectively (81.. 0.80%), respectively, (81.). 93% + 3.06%), (75.23% + 1.13%) and (70.66% + 5.63%), and the differences in each group were statistically significant (P0.05). After Nano-Cd S with 10 and 20 mg. L~ (-1) of 24 h, the intracellular ROS content and cell apoptosis rate increased with the increase of the dose. When the concentration was 10 mg L~ (-1), the cell apoptosis rate was (6.26% + 0.44%). The activity of SOD and CAT and the content of MDA in each group were increased. The activity and MDA content of SOD and CAT in the 20 mg. L~ (-1) group were higher than that of the 10 mg. L~ (-1).
