西蘭花莖葉多肽的酶法提取及其增強免疫力功能研究

【摘要】:本文對西蘭花莖葉多肽進行酶法提取以篩選增強免疫力功能活性強的組分。從西蘭花莖葉中提取粗蛋白后,首先經(jīng)酶法制備西蘭花多肽,然后采用Valflow50超濾、Sephadex G-15進行分離純化,得到分子量為310~2600 Da的西蘭花小肽,并對其進行增強免疫力功能的動物實驗研究,分別以100、50、25 mg/kg·bw 3個劑量的西蘭花多肽提取物對小鼠連續(xù)灌胃30 d,然后分別測定小鼠的體液免疫功能、細胞免疫功能和單核-巨噬細胞功能。結(jié)果表明,高劑量組(100 mg/kg)西蘭花多肽提取物對小鼠的淋巴細胞轉(zhuǎn)化能力(0.225±0.040)與小鼠的左右耳片重量差異(18.19±2.94)mg均顯著高于空白對照組(p0.05)。西蘭花多肽提取物對正常小鼠碳粒廓清功能有非常明顯的促進作用,碳廓清指數(shù)分別為(0.126±0.052)和(0.136±0.022),高、中劑量能顯著(p0.05)增加小鼠非特異免疫功能。以上結(jié)果表明西蘭花多肽提取物具有增強免疫力的功能,此研究可為西蘭花的深度開發(fā)和綜合利用提供基礎(chǔ)數(shù)據(jù)和科學參考。
[Abstract]:In this paper, broccoli stem and leaf polypeptides were extracted by enzymatic method to screen the components with strong immune function. After extracting crude protein from the stems and leaves of broccoli, broccoli peptides were prepared by enzymatic method, and then purified by Valflow50 ultrafiltration Sephadex G-15. The small peptides of broccoli with molecular weight of 310 ~ 2600Da were obtained. The mice were perfused with three doses of Broccoli polypeptide extract of 100m 50g 25 mg/kg BW for 30 days, then the humoral immune function of mice was measured. Cellular immune function and monocyte-macrophage function. The results showed that the lymphocyte transformation ability of high dose group was significantly higher than that of control group (0.225 鹵0.040) and the weight difference of left and right ear slices of mice (18.19 鹵2.94)mg) than that of control group (P 0.05). The polypeptide extract of broccoli significantly promoted the clearance of carbon particles in normal mice. The carbon clearance index was 0.126 鹵0.052) and 0.136 鹵0.022, respectively. The medium dose could significantly increase the non-specific immune function of mice. The above results indicate that the extract of broccoli polypeptide has the function of enhancing immunity. This study can provide basic data and scientific reference for the further development and comprehensive utilization of broccoli.
