氟鋁聯(lián)合持續(xù)暴露對二代仔鼠神經(jīng)生理發(fā)育及海馬結(jié)構(gòu)的影響

【摘要】:目的探討氟鋁聯(lián)合持續(xù)暴露對二代仔鼠神經(jīng)生理發(fā)育和海馬結(jié)構(gòu)的影響。方法 SD孕鼠36只,隨機分為9組(每組4只):對照組和單獨氟化鈉(100、200 mg/L)、三氯化鋁(500、1 000 mg/L)染毒組及氟化鈉+三氯化鋁聯(lián)合染毒組。從母鼠妊娠第0天至二代仔鼠出生第60天(PND60),自由飲水染毒。每組一代仔鼠于PND90(性成熟)隨機選取6只(雌雄比為2∶1)合籠。每組二代仔鼠出生后,隨機選取8只(雌雄各半),觀察神經(jīng)生理發(fā)育指標;PND60,收集12 h尿液,處死后留取大腦,離子選擇電極法、電感耦合等離子體發(fā)射光譜法分別測定尿和腦中氟、鋁;HE染色觀察海馬變化。結(jié)果與對照組比較,各染毒組腦重系數(shù)、開眼和出牙未見差異;高氟組、低氟低鋁組和高氟低鋁組平面翻正反射時間延長,高劑量氟、鋁及氟鋁聯(lián)合組二代仔鼠懸崖回避反射時間延長(P0.05)。HE染色顯示高劑量氟、鋁及氟鋁聯(lián)合組CA3區(qū)出現(xiàn)病理改變。結(jié)論高劑量氟、鋁及氟鋁聯(lián)合可能致二代仔鼠的神經(jīng)生理發(fā)育及海馬損傷。
[Abstract]:Objective to investigate the effects of combined fluoride and aluminum exposure on neurophysiological development and hippocampal formation in second generation rats.Methods Thirty-six pregnant SD rats were randomly divided into 9 groups (4 rats in each group: control group, 100 200 mg / L sodium fluoride alone group, 1 000 mg / L aluminum chloride 500 mg / L group) and sodium fluoride trichloride combined group.From the first day of pregnancy to the 60th day of birth of the second generation, the mice were exposed to the free drinking water.Six offspring (2: 1 female / male) were randomly selected from each group in PND 90 (sexual maturity).Eight rats (half male and half female) were randomly selected from each group after birth. The neurophysiological development index (PND60) was observed. Urine samples were collected for 12 hours. The brain was collected after death and the brain was retained by ion selective electrode method.Inductively coupled plasma emission spectrometry (ICP-AES) was used to detect fluoride in urine and brain respectively.Results compared with the control group, there was no difference in brain weight coefficient, open eye and teeth in each exposure group, and the time of positive reflex was prolonged in the high fluorine group, low fluoride low aluminum group and high fluorine low aluminum group, and high dose fluoride.The cliff avoidance reflex time of the second generation rats in the combined group of aluminum and fluorine was prolonged (P 0.05). He staining showed high dose of fluorine, and the pathological changes in the CA3 region of the group of aluminum and fluoro aluminum.Conclusion the combination of high dose fluoride, aluminum and aluminum fluoride may induce neurophysiological development and hippocampal injury in second generation rats.
