納米羥基磷灰石對3種正常組織細胞的DNA損傷作用研究

【摘要】:目的探討納米羥基磷灰石(HA-NPs)對3種正常組織細胞的DNA損傷作用,為HA-NPs毒性機制提供實驗依據(jù)。方法以正常人肝細胞(L02)、肺支氣管上皮細胞(HBE)和小鼠成骨細胞(MC3T3)為研究對象,以不同濃度HA-NPs染毒細胞24 h,采用MTT比色法檢測細胞毒性,同時用彗星實驗和熒光探針法分別檢測HA-NPs對細胞的DNA損傷和活性氧水平。結果 HA-NPs在較低濃度(0~400μg/ml)時促進3種細胞的生長,在大于400μg/ml時則抑制細胞生長。隨著HA-NPs濃度的增高,3種細胞的拖尾細胞數(shù)、尾部DNA百分比、Olive尾距(OTM)均逐漸增加,與對照組比較差異有統(tǒng)計學意義(P0.05)。3種細胞的活性氧熒光強度均隨HA-NPs濃度增加而增加,相較于對照組差異有統(tǒng)計學意義(P0.05)。結論 HA-NPs可引起3種受試細胞DNA損傷,其機制可能與HA-NPs誘導細胞產(chǎn)生活性氧有關。
[Abstract]:Objective to investigate the DNA damage effect of HA-NPs on three kinds of normal tissue cells, and to provide experimental evidence for the mechanism of HA-NPs toxicity. Methods normal human hepatocytes (L02), lung bronchial epithelial cells (HA-NPs) and mouse osteoblasts (MC3T3) were used as subjects. The cytotoxicity of cells exposed to different concentrations of HA-NPs for 24 h was detected by MTT colorimetry. At the same time, comet assay and fluorescence probe method were used to detect DNA damage and reactive oxygen species level of HA-NPs. Results HA-NPs promoted the growth of three kinds of cells at a low concentration of 400 渭 g / ml. The cell growth was inhibited when the concentration of HA-NPs was greater than 400 渭 g / ml. With the increase of HA-NPs concentration, the percentage of tail DNA increased gradually. Compared with the control group, the fluorescence intensity of reactive oxygen species increased with the increase of HA-NPs concentration, and the difference was statistically significant compared with the control group. Conclusion HA-NPs can cause DNA damage in three kinds of cells. The mechanism may be related to the production of reactive oxygen species (Ros) induced by HA-NPs.
