山楂黃烷醇延緩衰老活性及其作用機(jī)制研究

【摘要】:人口老齡化是我國(guó)等多個(gè)國(guó)家都在面臨的嚴(yán)峻問(wèn)題,因而衰老及其機(jī)理研究受到了廣泛的重視。本文依托衰老的自由基學(xué)說(shuō)、通過(guò)體內(nèi)試驗(yàn)對(duì)山楂黃烷醇(MF)的延緩衰老活性和作用機(jī)制進(jìn)行了探索,為進(jìn)一步開發(fā)保健食品新資源提供依據(jù)。 研究方法: (1)MF延緩果蠅衰老活性研究。利用果蠅生存實(shí)驗(yàn)?zāi)P陀^察受試物對(duì)雌、雄果蠅壽命參數(shù)——平均壽命、最高壽命和累計(jì)死亡曲線的影響。并對(duì)飼養(yǎng)30d的雌、雄果蠅進(jìn)行生化指標(biāo)檢測(cè),評(píng)估受試物在果蠅體內(nèi)的抗氧化活性。 (2)MF對(duì)D-半乳糖誘導(dǎo)小鼠過(guò)氧化損傷的拮抗作用。在對(duì)小鼠皮下注射D-半乳糖以制造過(guò)氧化損傷模型的過(guò)程中,同時(shí)灌胃給予受試物,連續(xù)49d;以小鼠血清和臟器的抗氧化酶(SOD、GSH-Px)活性、脂質(zhì)過(guò)氧化產(chǎn)物(MDA)含量及肝、腦衰老指示酶(MAO)活性為檢測(cè)指標(biāo),評(píng)估受試物對(duì)D-半乳糖誘導(dǎo)小鼠過(guò)氧化損傷的保護(hù)作用和預(yù)防衰老作用。 (3)MF對(duì)衰老模型小鼠機(jī)體抗氧化活性的影響。首先通過(guò)皮下注射D-半乳糖的方式復(fù)制衰老小鼠模型,造模成功后,灌胃給予受試物,連續(xù)35d;以衰老小鼠血清和臟器的抗氧化酶(SOD、GSH-Px)活性、脂質(zhì)過(guò)氧化產(chǎn)物(MDA)含量及肝、腦衰老指示酶(MAO)活性、腦ATP酶(Na+K+ATPase)為檢測(cè)指標(biāo),考察受試物對(duì)老齡動(dòng)物機(jī)體抗氧化水平的改善和恢復(fù)作用。 研究結(jié)果: (1)MF延緩果蠅衰老活性。MF可以提高雌、雄果蠅平均壽命和最高壽命,并推遲半數(shù)死亡時(shí)間;0.2%和0.8%劑量組具有顯著的延壽作用,且整體而言對(duì)雌果蠅效果更好;上述兩劑量組對(duì)雌果蠅的平均壽命延長(zhǎng)率分別為6.6%和7.2%,最高壽命延長(zhǎng)率分別為5.8%和12.1%,對(duì)雄果蠅的平均壽命延長(zhǎng)率分別5.3%和5.8%,最高壽命延長(zhǎng)率分別為和11.7%和10.2%。MF也可提升果蠅體內(nèi)SOD活力和降低MDA水平,0.2%劑量組效果最好,,對(duì)雌果蠅效果更好。MF的延壽作用可能與其抗氧化活性有關(guān)。 (2)MF對(duì)D-半乳糖誘導(dǎo)小鼠過(guò)氧化損傷的拮抗作用。在造模的同時(shí)給予MF可以拮抗D-半乳糖誘導(dǎo)的小鼠體內(nèi)過(guò)氧化損傷過(guò)程,不同劑量MF及陽(yáng)性對(duì)照物(100mg/kg.bw維生素C)在不同樣本中活性不同。從抗氧化指標(biāo)和衰老指示酶分析,60mg/kg.bw的MF有局部的抗氧化和預(yù)防衰老活性,120mg/kg.bw的MF則有較全面的抗氧化和延緩衰老活性,60~120mg/kg.bw對(duì)主要臟器中MDA的清除效果要優(yōu)于陽(yáng)性對(duì)照,而陽(yáng)性對(duì)照組對(duì)血清各指標(biāo)的效果較好,其它指標(biāo)MF與陽(yáng)性對(duì)照組各有千秋。 (3)MF對(duì)衰老模型小鼠機(jī)體抗氧化活性的影響。研究表明,不同劑量的MF可不同程度的減輕衰老模型小鼠的老化狀態(tài)、改善其抗氧化水平。90~180mg/kg.bw的MF在衰老動(dòng)物血清和臟器內(nèi)具有抗氧化活性,并可提升腦Na+K+ATPase水平,且高劑量效果更好、更全面;60mg/kg.bwMF可降低腦MAO活性,90~180mg/kg.bwMF可降低肝MAO活性。MF抗衰老作用的最佳作用條件還有待進(jìn)一步探索。
[Abstract]:In this paper , the mechanism of aging and its mechanism of aging and its mechanism are studied in this paper . Based on the theory of free radical in aging , the mechanism of delaying aging and the mechanism of action of hawthorn flavanol ( MF ) is explored through in vivo experiments , which provides the basis for further development of new resources for health food . Study method : ( 1 ) The effects of the test article on the life parameters , mean life , maximum life and cumulative death curve of female and male fruit fly were observed by using the survival experiment model of fruit fly . ( 2 ) The effect of MF on the peroxidation damage induced by D - galactose in mice was studied . In the process of producing peroxide injury model by subcutaneous injection of D - galactose , the activity of antioxidant enzymes ( SOD , GSH - Px ) , lipid peroxidation products ( MDA ) and liver and brain aging indicator enzyme ( MAO ) were measured . ( 3 ) The effects of MF on anti - oxidation activity of aging model mice were studied . First , the aging mice model was duplicated by subcutaneous injection of D - galactose . After successful molding , the test article was injected into the stomach for 35 days . The activities of antioxidant enzymes ( SOD , GSH - Px ) , lipid peroxidation products ( MDA ) and liver , brain aging indicator enzyme ( MAO ) activity and brain ATP enzyme ( Na + K + ATPase ) were measured by subcutaneous injection of D - galactose . Results of the study : The results showed that MF could increase the average life expectancy and the maximum life of the female and male fruit fly , postpone the half dead time ; 0.2 % and 0.8 % of the dose groups had significant life prolonging effects . The average life extension rate of the two groups were 6 . 6 % and 7 . 2 % respectively . ( 2 ) The effect of MF on peroxidation damage induced by D - galactose in mice induced by D - galactose . MF and positive control ( 100mg / kg . bw vitamin C ) had different activity in different samples . ( 3 ) The effect of MF on anti - oxidation activity of aging model mice was studied .
