【摘要】：建立牛乳制品中青霉素酶Alphalisa的檢測方法。將青霉素酶進(jìn)行生物素化,與樣品中的青霉素酶競爭結(jié)合鼠源性青霉素酶單克隆抗體,利用Alphalisa反向競爭的反應(yīng)模式,對牛乳中的青霉素酶實(shí)施檢測。建立的Alphalisa檢測方法能特異性識別樣品中存在的青霉素酶,方法的線性范圍在2.5~500 IU/m L;檢出限和定量限分別為1.6 IU/m L和5 IU/m L;批間批內(nèi)變異度均小于10%。結(jié)果表明,該方法能夠替代傳統(tǒng)酶聯(lián)免疫吸附實(shí)驗(yàn)方法,成為青霉素酶的實(shí)驗(yàn)室篩查或檢測方法。
[Abstract]:A method for the detection of penicillin Alphalisa in bovine dairy products was established. The penicillin enzyme was biotinylated to compete with the penicillin enzyme in the sample and combined with the monoclonal antibody against the mouse penicillin enzyme. The reverse competition reaction mode of Alphalisa was used. The penicillin enzyme in milk was detected. The established Alphalisa method can specifically identify the penicillin enzyme in the sample. The linear range of the method is 2.5 IU/m / L, the detection limit and quantitative limit are 1.6 IU/m / L and 5 IU/m / L, respectively, and the intra-assay variability is less than 10%. The results show that this method can replace the traditional enzyme linked immunosorbent assay (Elisa). Become a laboratory screening or detection method for penicillin.
