黑龍江地區(qū)典型人群酸菜中亞硝酸鹽暴露的調(diào)查研究

【摘要】:目的:通過調(diào)查黑龍江地區(qū)酸菜攝入水平及酸菜、泡菜中亞硝酸鹽的含量,了解該地區(qū)人群膳食酸菜、泡菜攝入亞硝酸鹽的情況,并進(jìn)一步探討亞硝酸鹽攝入與人體產(chǎn)生氧化應(yīng)激的關(guān)系。用流式細(xì)胞術(shù)檢測(cè)膳食酸菜及泡菜攝入亞硝酸鹽人群血漿中IL-17A、IFN-γ、TNF、IL-10、IL-6、IL-4、IL-2等細(xì)胞因子以及趨化因子CXCL10水平,探討膳食酸菜及泡菜而攝入亞硝酸鹽水平與人體炎性因子的關(guān)系。方法:通過黑龍江牡丹江市西安區(qū)海南鄉(xiāng)兩個(gè)村的典型人群調(diào)查,選擇80人作為調(diào)查對(duì)象,分別在12月初和2月底收集對(duì)象的一天的膳食酸菜、泡菜攝入樣本,進(jìn)行一般人口學(xué)調(diào)查,并收集兩次的血液樣本。通過GB 5009.33—2010測(cè)定膳食酸菜、泡菜中亞硝酸鹽的含量,結(jié)合膳食酸菜及泡菜的攝入計(jì)算膳食亞硝酸鹽的攝入量;利用比色法測(cè)定調(diào)查對(duì)象血漿中的亞硝酸鹽含量;利用比色法測(cè)定血漿中CAT、SOD的含量;采用流式細(xì)胞細(xì)胞術(shù)(CBA)檢測(cè)各組血漿中IL-17A、IFN-γ、TNF、IL-10、IL-4、IL-2等細(xì)胞因子水平,采用ELISA雙抗體夾心法檢測(cè)血漿中CXCL10水平。根據(jù)這種攝入的四分位數(shù)間距進(jìn)行分組,以下四分位數(shù)間距作為參比組(對(duì)照組),第1-4組分別為20、20、20、20。檢測(cè)并比較高攝入組與對(duì)照組人體血漿中CAT、SOD的活性水平。比較高攝入與對(duì)照組人體血漿中IL-17A、IFN-γ、TNF、IL-10、IL-4、IL-2等細(xì)胞因子及趨化因子CXCL10水平。結(jié)果:描述調(diào)查地區(qū)典型人群的一般人口學(xué)特征,第一次調(diào)查膳食酸菜攝入為195.98±79.32g/天/人,第二次調(diào)查膳食酸菜攝入為178.89±49.55g/天/人。第一次收集到70份酸菜樣品,第二次收集到66份酸菜樣品。共檢測(cè)136份樣品中亞硝酸鹽的檢出率為70.6%,超標(biāo)率為0.7%。第一次調(diào)查血漿樣品中亞硝酸鹽的平均濃度為13.79±11.27 uM,第二次調(diào)查收集到的80份血漿樣本中亞硝酸鹽的平均濃度為11.67±6.31 uM。以調(diào)查對(duì)象亞硝酸鹽攝入四分位間距進(jìn)行分組,1-4組分別亞硝酸鹽攝入分別為:82.98±75.86ug、273.77±33.81ug、405.27±39.63ug、723.33±147.95ug。膳食酸菜及泡菜后,人體血漿中亞硝酸鹽含量基本穩(wěn)定。膳食酸菜及泡菜后,人體血漿中中CAT活力、CXCL10水平趨于穩(wěn)定,SOD活力有一定的抑制作用。膳食酸菜及泡菜后人體血漿中IL-17A、IFN-γ、TNF、IL-10、IL-6、IL-4等細(xì)胞因子水平有所下降。結(jié)論:被調(diào)查地區(qū)酸菜、泡菜攝入量調(diào)查顯示,典型人群膳食酸菜及泡菜中亞硝酸鹽在國(guó)家標(biāo)準(zhǔn)限定的范圍內(nèi)。不同酸菜消費(fèi)者血液含量變化無差別。膳食酸菜及泡菜攝入亞硝酸鹽后,對(duì)SOD活力有一定的抑制作用。本試驗(yàn)研究膳食不同酸菜消費(fèi)者體內(nèi)炎性因子發(fā)生變化與亞硝酸鹽的攝入量存在一定相關(guān)性,隨著膳食酸菜及泡菜中亞硝酸鹽總攝入水平的增高,人體血漿中IL-17A、TNF、IL-10、IL-6、IL-4等細(xì)胞因子水平有所下降。
[Abstract]:Objective: to investigate the intake level of pickled vegetables and nitrite in pickles and pickles in Heilongjiang province, and to understand the intake of nitrite in pickled vegetables and pickles. The relationship between nitrite intake and oxidative stress in human body was further investigated. IL-17A- IFN- 緯 TNF- 偽 in plasma was detected by flow cytometry. IL-10, IL-6, IL-4, IL-2, and chemokine CXCL10. To explore the relationship between the levels of nitrite intake and human inflammatory factors in pickled vegetables and pickles. Methods: 80 people were selected from two villages of Hainan Township, Mudanjiang District, Heilongjiang Province. Samples of dietary pickles and pickles were collected in early December and the end of February for general demography. The contents of nitrite in pickled vegetables and pickles were determined by GB5009.33-2010. The intake of nitrite was calculated by combining the intake of pickled vegetables and pickles. The content of nitrite in plasma was determined by colorimetry. The content of SOD in plasma was determined by colorimetry. The levels of IL-17A- IFN- 緯 TNFN- 緯 TNFN- 10, IL-4, IL-4 and IL-2 in plasma of each group were detected by flow cytometry (CBA). Plasma CXCL10 levels were measured by ELISA double antibody sandwich method. The quartile spacing was grouped according to the quartile spacing of the intake, and the following quartile spacing was used as the reference group (control group). The levels of CAT in human plasma were detected and compared between the high intake group and the control group. The activity level of SOD was compared with that in the control group. The level of IL-17An- 緯 -TNFN- 緯 TNFN- 10 IL-10 IL-4 was compared with that in the control group. IL-2 and other cytokines and chemokine CXCL10 levels. Results: the general demographic characteristics of typical population in the investigated area were described. The first dietary intake of pickled vegetables was 195.98 鹵79.32g / day per person. The dietary intake of pickled vegetables was 178.89 鹵49.55g / day per person in the second survey. 70 samples of pickled vegetables were collected for the first time. 66 pickled vegetable samples were collected for the second time. The detection rate of nitrite was 70.6%. The average concentration of nitrite in plasma samples was 13.79 鹵11.27 UM for the first time. The average nitrite concentration of 80 plasma samples collected in the second survey was 11.67 鹵6.31 UM.The nitrite intake intervals were divided into groups. In the 1-4 group, the nitrite intake was:: 82.98 鹵75.86ugn 273.77 鹵33.81ugn 405.27 鹵39.63ug respectively. 723.33 鹵147.95ug.After the diet of pickled vegetables and pickles, the content of nitrite in human plasma was basically stable, and the activity of CAT in human plasma was observed after dietary pickled vegetables and pickles. The level of CXCL10 tended to stabilize and the activity of sod was inhibited to a certain extent. The plasma IL-17A IFN- 緯 TNFN- 10 IL-6 was found in the plasma of pickled vegetables and pickled vegetables. Conclusion: the intake of pickled vegetables and pickles in the investigated area was investigated. Dietary nitrite of pickled vegetable and pickled vegetable in typical population was within the limit of national standard. There was no difference in blood content of different pickled vegetable consumers. After ingesting nitrite in dietary pickled vegetable and pickled vegetable. SOD activity was inhibited to some extent. In this experiment, the changes of inflammatory factors in different pickled vegetable consumers were correlated with the intake of nitrite. With the increase of nitrite intake in pickled vegetables and pickled vegetables, the levels of cytokines such as IL-17, TNFN, IL-10, IL-6 and IL-4 in human plasma decreased.
