【摘要】：目的觀察鄰苯二甲酸(2-乙基已基)酯(DEHP)染毒90 d對大鼠精子運動能力的毒性作用。方法將健康清潔級SD雄性大鼠40只隨機分為對照組和3個DEHP染毒組(5、50、500 mg/kg),進行90 d經(jīng)口染毒;取睪丸、附睪稱重,并用擴散法收集大鼠附睪尾精子,應(yīng)用計算機輔助精子分析系統(tǒng)(CASA)對精子的運動參數(shù)進行檢測。結(jié)果 5 mg/kg DEHP組大鼠精子直線性(LIN)為(44.00±2.52)%,明顯低于對照組(50.00±2.71)%(P0.05);500 mg/kg DEHP組大鼠精子鞭打頻率(BCF)為(5.8±4.2)Hz,明顯低于對照組(10.4±1.7)Hz(P0.05);與對照組比較,500 mg/kg DEHP組大鼠精子的平均路徑速度(VAP)、直線運動速度(VSL)、曲線運動速度(VCL)、精子頭側(cè)擺幅度(ALH)、BCF明顯下降,差異均有統(tǒng)計學(xué)意義(均P0.05);隨著DEHP染毒濃度增加,大鼠精子各項運動參數(shù)逐漸降低,精子的運動能力下降。結(jié)論 DEHP對大鼠附睪精子的運動能力有直接毒性作用。
[Abstract]:Objective to observe the content of 2-ethylhexyl phthalate (DEHP). Methods Forty SD male rats of clean grade were randomly divided into two groups: control group and 3 DEHP groups. 50,500 mg / kg, administered orally for 90 days; Testis were taken and epididymis was weighed and sperm was collected from rat epididymal tail by diffusion method. The sperm motility parameters were measured by computer aided sperm analysis system (CASA). Results the sperm linear line in 5 mg/kg DEHP group was (. 44.00 鹵2.52%. It was significantly lower than that in the control group (50.00 鹵2.71). The frequency of sperm whipping in the 500 mg/kg DEHP group was 5.8 鹵4.2 Hz, which was significantly lower than that in the control group (10.4 鹵1.7). Compared with the control group, the average path velocities of spermatozoa, linear motion velocities and curvilinear motion velocities were observed in the #number0# mg/kg DEHP group. The amplitude of head and side pendulum of spermatozoa decreased significantly (P 0.05). With the increase of the concentration of DEHP, the motility of sperm decreased and the motility of sperm decreased. Conclusion DEHP has a direct toxic effect on the motility of rat epididymal spermatozoa.
