新生期雙酚A染毒對小鼠睪丸Boule基因表達(dá)的長期影響

【摘要】:目的探討新生小鼠雙酚A染毒對小鼠睪丸結(jié)構(gòu)及Boule基因表達(dá)的長期影響及機(jī)制。方法將12只清潔級妊娠ICR母鼠,待分娩后隨機(jī)分為空白對照組、陰性對照組(玉米油)和雙酚A(100μg/kg)組。仔鼠從出生第1天開始按4μL/g BW頸背部皮下注射藥物,直到出生第21天停止注射,出生第35和70天處死取材。蘇木精-伊紅染色觀察小鼠睪丸組織形態(tài)學(xué)變化,采用RT-PCR法測定睪丸Boule mRNA表達(dá)量變化,免疫熒光和Western blot法檢測睪丸內(nèi)Boule蛋白的表達(dá)水平。結(jié)果與空白對照組、陰性對照組相比,出生第35和70天的雙酚A組睪丸曲細(xì)精管直徑與生精上皮厚度顯著減小,管腔顯著增大(P0.05),睪丸中Boule mRNA和蛋白表達(dá)水平均顯著降低(P0.05,P0.01)。結(jié)論新生期雙酚A染毒可通過降低睪丸組織內(nèi)Boule mRNA和蛋白質(zhì)的表達(dá)水平進(jìn)而影響睪丸發(fā)育。
[Abstract]:Objective to investigate the long-term effect and mechanism of bisphenol A (BPA) exposure on testis structure and Boule gene expression in neonatal mice. Methods Twelve pregnant ICR mice of clean grade were randomly divided into blank control group after delivery. In the negative control group (corn oil) and bisphenol A 100 渭 g / kg group, the rats were injected subcutaneously with 4 渭 L / g BW on the first day of birth until the 21st day of birth. On the 35th and 70th day of birth, the specimens were sacrificed. The morphological changes of testis were observed by hematoxylin and eosin staining, and the expression of Boule mRNA in testis was determined by RT-PCR method. The expression of Boule protein in testis was detected by immunofluorescence and Western blot. The diameter of testicular seminiferous tubules and the thickness of seminiferous epithelium in group A were significantly decreased and the lumen was significantly increased (P0.05) on the 35th and 70th day of birth. The expression of Boule mRNA and protein in testis decreased significantly (P0.05). Conclusion BPA exposure may affect testicular development by decreasing the expression of Boule mRNA and protein in testis.
