目的探討雌馬酚對(duì)PC12細(xì)胞缺氧/復(fù)氧損傷的保護(hù)作用及機(jī)制。方法以PC12細(xì)胞為研究對(duì)象,構(gòu)建體外神經(jīng)元缺氧/復(fù)氧損傷模型,用雌馬酚進(jìn)行干預(yù)。MTT法檢測(cè)細(xì)胞活力,比色法測(cè)定細(xì)胞培養(yǎng)上清液中乳酸脫氫酶（LDH）活性和丙二醛（MDA）含量,免疫細(xì)胞化學(xué)法檢測(cè)活性氧（ROS）含量,Western blotting檢測(cè)缺氧/復(fù)氧損傷的PC12細(xì)胞內(nèi)NADPH氧化酶2（gp91phox）和磷酸化Src（p-Src）激酶的表達(dá)。結(jié)果雌馬酚可有效減輕缺氧/復(fù)氧引起的PC12細(xì)胞損傷,降低缺氧/復(fù)氧損傷細(xì)胞培養(yǎng)上清液中LDH的活性和MDA的含量,明顯抑制ROS的生成及gp91^phox和p-Src的表達(dá)。結(jié)論雌馬酚通過抑制p-Src激酶和gp91^phox表達(dá),減少ROS生成,從而對(duì)PC12細(xì)胞缺氧/復(fù)氧損傷產(chǎn)生保護(hù)作用。
