ATRA衍生物Fenretinide對(duì)肝癌增殖、遷移的影響及其機(jī)制的研究

The effect of fenretinide on the morphology of HepG2 cellsanges in cell morphology were observed when HepG2 cells were treated with trations of fenretinide or ATRA for 48 h, and cells were photographed using Leica DMcope (magnification, x100).enretinide 和 ATRA 作用 48 h,光鏡下觀察 SMMC-7721 細(xì)胞發(fā)現(xiàn):對(duì)照和溶劑 DMSO 對(duì)照)如下圖 6,細(xì)胞生長(zhǎng)密集,光澤度好;5 μM 和inide 處理組(如下圖 6)細(xì)胞密度均較對(duì)照組明顯降低,生長(zhǎng)稀疏,,細(xì)胞核出現(xiàn)腫脹、破裂等,細(xì)胞失去貼壁生長(zhǎng)特性,高濃度組現(xiàn)時(shí),細(xì)胞液顏色亦有改變,光鏡下拍出的背景顏色區(qū)別較大。15 μM(如下圖 6)細(xì)胞亦有減少,無(wú)堆疊生長(zhǎng),但形態(tài)改變不顯著,5 μM(如下圖 6)與對(duì)照組相比,形態(tài)和密度改變均不顯著。

The effect of fenretinide on the morphology of SMMC-7721 cellse changes in cell morphology were observed when SMMC-7721 cells were treated with diffcentrations of fenretinide or ATRA for 48 h, and cells were photographed using Leica DMI3croscope (magnification, x100).3 Fenretinide 抑制 HepG2、SMMC-7721 細(xì)胞遷移細(xì)胞劃痕實(shí)驗(yàn)顯示 fenretinide 以劑量依賴方式抑制 HepG2 細(xì)胞的遷移,如 7。24 h 遷移率分別為:5 μM,0.22±0.04,10 μM,0.19±0.05;48 h 遷移率分:5 μM,0.51±0.07,10 μM,0.32±0.01。而細(xì)胞對(duì)照組的遷移率 24 h 是 0.45±0 h 是 0.74±0.05。統(tǒng)計(jì)結(jié)果顯示 fenretinide 在 24 h 和 48 h 可顯著抑制 HepG的遷移(p㩳0.05),,如下圖 7B。ATRA 僅高濃度 10 μM 組有抑制 HepG2 細(xì)胞
