度他雄胺對大鼠附睪Claudin1和β-catenin表達的影響

【摘要】:目的:通過度他雄胺對大鼠附睪上皮連接關(guān)鍵蛋白Claudin1和β-catenin表達影響的研究,探討度他雄胺抑制生育的可能分子機制,為男性不育治療和男性避孕提供附睪靶點。方法:16只SD雄性大鼠(3月齡,250~300 g),設(shè)度他雄胺40 mg/kg實驗組和陰性對照組,每組8只,灌胃給藥,1次/d,連續(xù)2周。給藥結(jié)束后計算機輔助精子分析雄鼠精子活力及形態(tài),計算交配雌鼠受孕率,ELISA法測定大鼠血清睪酮(T)和雙氫睪酮(DHT)濃度,透射電鏡分析附睪細胞緊密連接變化,RT-PCR分析實驗組和對照組附睪上皮連接蛋白Claudin1和β-catenin基因表達變化;免疫組化和免疫印跡分析其蛋白表達變化。結(jié)果:度他雄胺可以顯著抑制大鼠血清DHT濃度、精子活力和生育力,超微結(jié)構(gòu)顯示實驗組附睪上皮細胞緊密連接出現(xiàn)空隙,附睪腔液體積增多。RT-PCR、免疫組化和免疫印跡顯示度他雄胺可以顯著下調(diào)Claudin1和β-catenin的表達。結(jié)論:度他雄胺可以顯著抑制雄性大鼠生育,這可能與其降低大鼠血清DHT水平,進而抑制附睪上皮連接關(guān)鍵基因Claudin1和β-catenin表達和破壞附睪上皮緊密連接有關(guān)。
