【摘要】：目的對安立生坦合成工藝的改進(jìn)進(jìn)行研究。方法以二苯甲酮為起始原料,經(jīng)過縮合、醇解、水解、拆分和取代反應(yīng)合成安立生坦。醇解采用鹽酸進(jìn)行反應(yīng),采用拆分劑鹽酸鹽和堿試劑通過"一鍋法"進(jìn)行拆分。結(jié)果優(yōu)化了醇解和拆分反應(yīng),避免了高毒性物質(zhì)的產(chǎn)生;優(yōu)化后,安立生坦的質(zhì)量分?jǐn)?shù)為99.7%,光學(xué)純度達(dá)99.8%,工藝的總收率為30.0%。結(jié)論安立生坦的合成工藝改進(jìn)后操作性好,條件溫和,避免產(chǎn)生基因毒性和其他高毒性物質(zhì),成本較低,更適于藥品的工業(yè)化生產(chǎn)。
[Abstract]:Objective to study the improvement of the synthesis process of Anlisheng Tan. Methods Anrisentam was synthesized from benzophenone by condensation, alcoholysis, hydrolysis, resolution and substitution. The alcoholysis was carried out by hydrochloric acid, and the split agent hydrochloric acid and alkali reagent were separated by one pot method. Results the alcoholysis and resolution reaction were optimized to avoid the production of high toxic substances, the mass fraction of Anrisentan was 99.7%, the optical purity was 99.8%, and the total yield of the process was 30.0%. Conclusion the improved synthetic process of Anrisentam has good maneuverability and mild conditions, avoids the production of genotoxicity and other high toxic substances, and has low cost, so it is more suitable for industrial production of drugs.
