HPLC法測定人血漿中伏立康唑的濃度

【摘要】:目的:建立一種快速、靈敏、準(zhǔn)確、穩(wěn)定的測定人血漿中伏立康唑濃度的方法,用于伏立康唑臨床治療藥物監(jiān)測。方法:采用高效液相色譜(HPLC)-紫外檢測法,以卡馬西平為內(nèi)標(biāo),乙腈蛋白沉淀法處理血漿樣品。色譜柱為Phecda C18,流動相為20mmol/L磷酸二氫鉀緩沖液(p H=6.0)-乙腈(50∶50,V/V),流速為1 ml/min,柱溫為40℃,檢測波長為255 nm,進(jìn)樣量為50μl。結(jié)果:伏立康唑和卡馬西平的保留時間分別為8.34和6.24 min;血漿中伏立康唑線性范圍為0.10~20.00μg/ml(r=0.999 5),定量下限為0.05μg/ml,日內(nèi)、日間RSD分別為≤1.57%、1.45%,低、中、高梯度濃度提取回收率在81.40%~128.29%之間。結(jié)論:該方法操作簡便,結(jié)果準(zhǔn)確,適用于伏立康唑的臨床治療藥物監(jiān)測。
