阿苯達(dá)唑納米脂質(zhì)體凍干粉的制備及性質(zhì)考察

【摘要】:目的:制備阿苯達(dá)唑納米脂質(zhì)體凍干粉并對(duì)其性質(zhì)進(jìn)行考察。方法:利用冷凍干燥法制備阿苯達(dá)唑納米脂質(zhì)體凍干粉,以粒徑、包封率聯(lián)合外觀、再分散性為指標(biāo),采用單因素試驗(yàn)聯(lián)合正交試驗(yàn)篩選凍干處方工藝?疾靸龈汕啊⒑笾|(zhì)體的形態(tài)學(xué)變化、粒徑、Zeta電位、水分含量、4℃下12個(gè)月的穩(wěn)定性。結(jié)果:采用外加凍干保護(hù)劑的總量為10%,其中葡萄糖-海藻糖-甘露醇配比為1.0∶1.0∶3.0,以速凍的方式,于-35℃冰箱預(yù)凍18 h,冷凍干燥48 h獲得凍干粉。與凍干前比較,凍干后脂質(zhì)體形態(tài)未發(fā)生明顯變化,可見(jiàn)清晰的磷脂雙分子層膜結(jié)構(gòu);凍干前、后脂質(zhì)體的粒徑分別為(208.63±1.04)、(223.04±2.02)nm,Zeta電位分別為(-15.6±0.04)、(-19.4±0.06)m V,包封率分別為(94.62±0.49)%、(91.10±0.46)%(n=3);與脂質(zhì)體比較,脂質(zhì)體凍干粉在4℃下12個(gè)月較穩(wěn)定。結(jié)論:成功制得阿苯達(dá)唑納米脂質(zhì)體凍干粉,其穩(wěn)定性優(yōu)于阿苯達(dá)唑納米脂質(zhì)體,凍干工藝可行。
