腎上腺髓質(zhì)素對(duì)大鼠肢體缺血再灌注后心肌損傷和細(xì)胞凋亡的影響

【摘要】:目的觀察大鼠肢體缺血再灌注后心肌細(xì)胞凋亡情況,探討外源性腎上腺髓質(zhì)素(ADM)對(duì)大鼠肢體缺血再灌注(LIR)后心肌損傷的保護(hù)機(jī)制。方法健康雄性Wistar大鼠30只,隨機(jī)分成對(duì)照組(Control組)、肢體缺血再灌注4 h組(LIR組)和ADM+LIR組(ADM組),每組10只。生化法檢測(cè)血漿心肌酶含量;比色法測(cè)定心肌組織丙二醛(MDA)和超氧化物歧化酶(SOD)含量的變化;免疫組織化學(xué)法檢測(cè)心肌組織凋亡蛋白酶Caspase-3蛋白表達(dá)情況;原位末端脫氧核糖核苷酸轉(zhuǎn)移酶標(biāo)記(TUNEL)技術(shù)檢測(cè)心肌凋亡細(xì)胞。結(jié)果與LIR組比較,心肌酶含量下降,心肌組織MDA含量減少,SOD活性增強(qiáng)(P0.01);心肌組織Caspse-3蛋白表達(dá)下降,心肌細(xì)胞凋亡指數(shù)明顯下降(P0.01)。結(jié)論 ADM可減輕大鼠LIR后心肌損傷,其機(jī)制可能與抑制心肌氧化應(yīng)激與細(xì)胞凋亡有關(guān)。
[Abstract]:Aim to observe the apoptosis of cardiomyocytes after limb ischemia-reperfusion in rats and to explore the protective mechanism of exogenous adrenomedullin (ADM) on myocardial injury after ischemia-reperfusion (LIR) in limbs of rats. Methods Thirty healthy male Wistar rats were randomly divided into three groups: control group (Control group), limb ischemia-reperfusion group (LIR group) and ADM LIR group (ADM group), with 10 rats in each group. Plasma myocardial enzyme content was measured by biochemical method, malondialdehyde (MDA) and superoxide dismutase (SOD) content were measured by colorimetric method, expression of apoptotic protease Caspase-3 protein was detected by immunohistochemical method. Cardiomyocyte apoptosis was detected by in situ terminal deoxynucleotidyl transferase labeled (TUNEL) technique. Results compared with LIR group, the content of myocardial enzyme decreased, the content of MDA decreased and the activity of SOD increased (P0.01), the expression of Caspse-3 protein decreased and the index of cardiomyocyte apoptosis decreased significantly (P0.01). Conclusion ADM can reduce myocardial injury after LIR, and its mechanism may be related to the inhibition of myocardial oxidative stress and apoptosis.
