甾體激素類藥物與細胞色素P450代謝酶系相互作用的研究進展

【摘要】:甾體激素主要包括腎上腺糖皮質(zhì)激素、雌激素、孕激素、雄激素和維生素D,臨床常用的甾體激素類藥物多為其衍生物或人工合成品。細胞色素P450(CYP450)代謝酶系在甾體激素的生物合成及代謝清除過程中起著關(guān)鍵的作用,而甾體激素也可通過對CYP450的誘導(dǎo)或抑制而影響其酶蛋白表達及代謝活性,二者之間的相互作用決定了臨床多種疾病(如腫瘤、炎癥或自身免疫性疾病)發(fā)生的易感性及藥物療效。本文對甾體激素類藥物與細胞色素P450代謝酶系相互作用的研究進展進行闡述。
[Abstract]:Steroids mainly include adrenocortical hormone, estrogen, progesterone, androgen and vitamin D. The steroids commonly used in clinic are mostly derivatives or synthetic products of steroids. Cytochrome P450 (CYP450) metabolizing enzymes play a key role in the biosynthesis and metabolic clearance of steroidal hormones. Steroid hormones can also affect the expression and metabolic activity of CYP450 by inducing or inhibiting cytochrome P450 metabolizing enzymes. The interaction between the two determines the susceptibility and drug efficacy of many clinical diseases, such as tumor, inflammation or autoimmune disease. The interaction between steroid hormones and cytochrome P450 metabolic enzymes was reviewed in this paper.
