環(huán)氧化酶1、P2Y1、GPⅠa及GPⅢa基因多態(tài)性與阿司匹林抵抗的相關(guān)性研究

【摘要】:目的探討環(huán)氧化酶1(COX-1)(A842G、C50T)、P2Y1(A1622G、C893T)、GPⅠa(C807T、G873A)及GPⅢa(T1565C)4個基因中7個位點(diǎn)的多態(tài)性與阿司匹林(ASP)抵抗(AR)的相關(guān)性。方法選取2011年10月—2013年12月武警浙江省總隊(duì)杭州醫(yī)院內(nèi)科住院的動脈粥樣硬化患者243例,測定患者血小板聚集(PA)率,并將患者分為AR組,ASP半抵抗(ASR)組,ASP敏感(AS)組。測定各組COX-1、P2Y1、GPⅠa及GPⅢa基因型。比較3組患者一般資料及COX-1、P2Y1、GPⅠa、GPⅢa基因多態(tài)性。結(jié)果 AR組28例,ASR組71例,AS組144例。3組患者的年齡、性別、吸煙率、高血壓發(fā)生率、總膽固醇、三酰甘油和低密度脂蛋白水平比較,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。3組患者糖尿病發(fā)生率比較,差異有統(tǒng)計(jì)學(xué)意義(P0.01);AS組患者糖尿病發(fā)生率低于AR組(P0.05)。3組間COX-1(842 AG)、COX-1(50 CT)、P2Y1(1622 AG)、GPⅠa(873 GA)、GPⅢa(1565 TC)頻率比較,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。3組間P2Y1(893 CT)和GPⅠa(807 CT)頻率比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05);AR組和ASR組P2Y1(893 CT)、GPⅠa(807 CT)頻率高于AS組(P0.01)。結(jié)論 P2Y1(C893T)、GPⅠa(C807T)等位基因的變異與AR具有一定程度的關(guān)聯(lián),為指導(dǎo)臨床用藥提供了參考依據(jù)。
