EGCG對大鼠三硝酸苯磺酸結(jié)腸炎Caspase-1表達及其相關(guān)細胞因子分泌的影響
[Abstract]:Aim: to observe the therapeutic effect of epigallocatechin gallate (EGCG) on TNBS colitis in rats and to explore its anti-inflammatory mechanism. Methods: the rat model of TNBS colitis was established by enema with TNBS/ ethanol solution. The rats were randomly divided into two groups: model group (25,50,75 mg / kg) and control group (control group). Each group was fed with 2ml according to the corresponding dose of EGCG, and the blank control group and the model group were given normal saline for 7 days. The disease activity index (DAI), colonic morphological injury index (CMDI), histopathological (HI) score (HI), malondialdehyde (MDA) content and myeloperoxidase (MPO) activity in colon tissue homogenate were observed, and the activity of myeloperoxidase (MPO) in colon tissue was measured. The expression of Caspase-1 in colon tissue was detected by immunohistochemical method, and the levels of IL-1 尾 and IL-18 in serum were detected by ELISA method. Results: compared with the model group, the general condition of rats was improved, the scores of DAI,CMDI and HI were decreased, and the activity of MPO and the content of MDA in colon tissue homogenate were also decreased in different degrees when the concentration of EGCG 75mg/kg was higher than that of the model group. Immunohistochemistry showed that the expression of Caspase-1 and the concentration of IL-1 尾 and IL-18 in the colon tissue were lower than those in the control group. Conclusion: EGCG can alleviate colon inflammation in rats with TNBS colitis, which may be related to EGCG inhibiting the expression of Caspase-1 in colon tissue and decreasing the levels of IL-1 尾 and IL-18 in serum.
