RP-HPLC法測定去甲斑蝥素齊墩果酸復(fù)合脂質(zhì)體的包封率

【摘要】:目的:建立測定去甲斑蝥素齊墩果酸復(fù)合脂質(zhì)體包封率的方法。方法:采用反相高效液相色譜法。色譜柱為Thermo Sycronis C18,流動(dòng)相為磷酸水溶液(p H=3.2)-甲醇(梯度洗脫),流速為1.0~1.7 ml/min,檢測波長為210 nm,柱溫為30℃,進(jìn)樣量為50μl。結(jié)果:去甲斑蝥素、齊墩果酸的質(zhì)量濃度分別在20~300、10~150μg/ml范圍內(nèi)與峰面積呈良好線性關(guān)系(r=0.999 8、0.999 7);精密度、穩(wěn)定性、重復(fù)性試驗(yàn)的RSD≤5.89%;去甲斑蝥素的平均回收率為99.15%~100.11%,RSD≤3.20%(n=5),齊墩果酸的平均回收率為99.22%~100.30%,RSD≤3.34%(n=5);去甲斑蝥素、齊墩果酸平均包封率分別為(48.01±2.34)%、(87.31±0.56)%(n=5)。結(jié)論:該方法法簡便、快速、可靠,可用于去甲斑蝥素齊墩果酸復(fù)合脂質(zhì)體包封率的測定。
