抗炎化合物HB0314在大鼠體內(nèi)的藥動學(xué)及絕對生物利用度研究

【摘要】:目的:建立大鼠血漿樣品中抗炎化合物HB0314的檢測方法,并研究其在大鼠體內(nèi)的藥動學(xué)特征。方法:采用超高效液相色譜-串聯(lián)質(zhì)譜法(UPLC-MS/MS),色譜柱為Waters Acquity UPLC~(TM)BEH C_(18)柱,流動相為0.1%甲酸水溶液(A)-甲醇(B),梯度洗脫(0~2 min,70%~90%B),流速為0.25 mL/min,柱溫為30℃,進樣量為5μL;采用電噴霧離子源,毛細管電壓為3 k V,離子源溫度為150℃,脫溶劑氣溫度為450℃,脫溶劑氣流量為600 L/h,錐孔氣流量為45 L/h,內(nèi)標為延胡索乙素。將12只大鼠隨機分成iv組和ig組,每組6只,分別iv、ig HB0314溶液5、10 mg/kg,分別于給藥前及給藥后5、15、30、60、120、240、360、480、600、720、1 440 min取頸靜脈血0.4 mL測定HB0314血藥濃度,采用DAS 2.0軟件計算藥動學(xué)參數(shù)并計算絕對生物利用度。結(jié)果:HB0314質(zhì)量濃度在1~1 000 ng/mL內(nèi)線性關(guān)系良好(r=0.995 5),定量下限為1 ng/mL;日內(nèi)、日間精密度、穩(wěn)定性的RSD≤8.45%(n=5);回收率為68.21%~90.29%(RSD≤11.20%,n=5);基質(zhì)效應(yīng)為82.63%~106.90%(RSD≤6.75%,n=5)。iv與ig給藥后,HB0314在大鼠體內(nèi)的AUC_(0-24 h)分別為(270.267±21.164)、(252.755±26.169)μg·h/L(n=6),t1/2z分別為(8.722±2.266)、(11.877±4.517)h(n=6),絕對生物利用度為56.97%。結(jié)論:本方法快速、簡便,可用于大鼠血漿中HB0314含量的測定。HB0314在大鼠體內(nèi)具有較高的絕對生物利用度,提示后期劑型設(shè)計可考慮將其制成口服劑。
